【摘要】： 目的:觀察去甲腎上腺素作用THP-1細(xì)胞、THP-1巨噬細(xì)胞和內(nèi)皮細(xì)胞后CD137、IL-6以及HMGB1基因表達(dá)的影響。 方法:對(duì)THP-1細(xì)胞、THP-1巨噬細(xì)胞和內(nèi)皮細(xì)胞給予應(yīng)激濃度的去甲腎上腺素刺激(劑量10μmol/L,時(shí)間1~24h),運(yùn)用逆轉(zhuǎn)錄-多聚酶鏈?zhǔn)椒磻?yīng)檢測(cè)CD137、IL-6和HMGB1 mRNA的表達(dá),酶聯(lián)免疫吸附試驗(yàn)檢測(cè)IL-6蛋白的表達(dá),Western印跡檢測(cè)CD137蛋白的表達(dá)。 結(jié)果: 10μmol/L去甲腎上腺素作用THP-1細(xì)胞、THP-1巨噬細(xì)胞和內(nèi)皮細(xì)胞1~24h后,其CD137、IL-6的mRNA和蛋白水平均上升,HMGB1 mRNA表達(dá)上升(p0.05)。 結(jié)論:應(yīng)激濃度的去甲腎上腺素能不同程度的上調(diào)THP-1細(xì)胞、THP-1巨噬細(xì)胞和內(nèi)皮細(xì)胞CD137、IL-6和HMGB1基因的表達(dá)。
[Abstract]:Objective: To observe the effect of norepinephrine on the expression of CD137, IL-6 and HMGB1 gene in THP-1 cells, THP-1 macrophages and endothelial cells.
Methods: THP-1 THP-1 cells, macrophages and endothelial cells to stress concentration of norepinephrine (dose 10 mol/L, time 1~24h, CD137) were detected by reverse transcriptase polymerase chain reaction, the expression of IL-6 and HMGB1 mRNA, the expression of IL-6 protein by enzyme-linked immunosorbent assay, the expression of CD137 protein was detected by Western blotting.
Results: the effect of 10 mol/L of norepinephrine on THP-1 cells, THP-1 macrophages and endothelial cells after 1~24h increased mRNA and protein levels of CD137, IL-6, and HMGB1 mRNA expression increased (P0.05).
Conclusion: the stress concentration of norepinephrine can increase the expression of CD137, IL-6 and HMGB1 genes in THP-1 cells, THP-1 macrophages and endothelial cells in varying degrees.
