【摘要】:目的 探討膽紅素(Bilirubin)對臍血T淋巴細(xì)胞功能的影響。 方法 選擇21例正常足月新生兒為研究對象,收集并分離臍血單個(gè)核細(xì)胞(CBMC),分別置于含有不同濃度膽紅素(0mg/dl,6mg/dl,9mg/dl,12mg/dl)的牛血清白蛋白(BSA/PBS)溶液中進(jìn)行預(yù)先孵育,洗滌后經(jīng)植物血凝素(PHA)刺激培養(yǎng)48小時(shí),采用丫錠橙/溴化乙錠(AB/EO)染色法測定細(xì)胞凋亡水平,間接免疫熒光法測定CBMC中CD25~+(IL-2受體)細(xì)胞百分率,酶聯(lián)免疫吸附試驗(yàn)(ELISA)檢測培養(yǎng)上清中IL-4及IFN-γ水平。 結(jié)果 ①膽紅素濃度0mg/dl組、6mg/dl組、9mg/dl組、12mg/dl組細(xì)胞凋亡率分別為(28.72±11.09)%,(32.44±12.72)%,(56.36±9.24)%,(61.49±8.93)%。12mg/dl組顯著高于6mg/dl組、Omg/dl組(q=12.55、14.16,p0.01),9mg/dl組亦顯著高于6mg/dl組、0mg/dl組(q=10.34、11.94,p0.01),而12mg/dl組與9mg/dl組間比較細(xì)胞凋亡水平差異無顯著性(q=2.22,p0.05),6mg/dl組與0mg/dl組比較細(xì)胞凋亡率亦無顯著性差別(q=1.60,p0.05)。②膽紅素濃度0mg/dl組、6mg/dl組、9mg/dl組、12mg/dl組CBMC體外經(jīng)PHA刺激后CD25陽性細(xì)胞百分率分別為(64.25±6.18)%,(67.28±4.87)%,(22.12±4.61)%,(19.89±6.72)%。12mg/dl組CD25陽性細(xì)胞百分率明顯低于6mg/dl組、0mg/dl組(q=38.35、35.90,p0.01),9mg/dl組亦明顯低于6mg/dl組、0mg/dl組(q=36.54、34.09,p0.01),但12mg/dl組與9mg/dl組間比較無顯著性差別(q=1.81,p0.05),且6mg/dl組與0mg/dl組比較CD25陽性細(xì)胞百分率亦無顯著性差別(q=2.45,p0.05)。③CBMC體外經(jīng)不同濃度膽紅素作用后經(jīng)PHA誘生IFN-γ水平12mg/dl組明顯低于6mg/dl組、0mg/dl組(q=38.17、41.95,p0.01),9mg/dl組亦明顯低于6mg/dl組、0mg/dl組(q=35.95、39.73,p0.01),6mg/dl組低于0mg/dl組,差異有統(tǒng)計(jì)學(xué)意義(q=3.78,p0.05),而12mg/dl組與9mg/dl組比較差異無顯著性(q=2.21,p0.05)。④各組培養(yǎng)上清中IL-4產(chǎn)生水平比較均無顯著性差異(p0.05)。⑤各組培養(yǎng)上清中IFN-γ/IL-4比值12mg/dl組明顯低于6mg/dl組、0mg/dl組(q=13.28、16.23,p0.01),9mg/dl組明顯低于6mg/dl組、0mg/dl組(q=12.49、15.44,p0.01),6mg/dl組低于0mg/dl組,差異有統(tǒng)計(jì)學(xué)意義(q=2.95,p0.05),但12mg/dl組與9mg/dl組比較差異無顯著性(q=0.79,p0.05)。⑥經(jīng)不同濃度膽紅素作用的CBMC經(jīng)48小時(shí)培養(yǎng)后,CD25陽性細(xì)胞百分率與凋亡細(xì)胞百分率呈顯著負(fù)相關(guān)(r=-0.75,p0.01),培養(yǎng)上清中IFN-γ水平、IFN-γ/IL-4比值亦與凋亡細(xì)胞百分率呈顯著負(fù)相關(guān)(r=-0.79、-0.69,p0.01),而培養(yǎng)上清中IL-4產(chǎn)生水平與細(xì)胞凋亡率無明顯相關(guān)性(r=-0.20,
[Abstract]:Objective to investigate the effect of bilirubin (Bilirubin) on the function of T lymphocyte in umbilical cord blood.
Methods 21 cases of normal full-term newborns as the research object, collecting and separating umbilical cord blood mononuclear cells (CBMC), were placed with different concentrations of bilirubin (0mg/dl, 6mg/dl, 9mg/dl, 12mg/dl) of bovine serum albumin (BSA/PBS) solution were pre incubated after washing by phytohemagglutinin (PHA) stimulated for 48 hours. Ingot acridine orange / ethidium bromide (AB/EO) staining to determine apoptosis level, determination of CBMC CD25~+ in indirect immunofluorescence (IL-2 receptor) cells, enzyme-linked immunosorbent assay (ELISA) and IL-4 IFN- gamma detection levels in supernatant of culture.
