【摘要】：Ⅰ型超敏反應(yīng)又稱速發(fā)型超敏反應(yīng),它的主要特征:發(fā)病快,機(jī)體出現(xiàn)生理功能性紊亂,有明顯個(gè)體差異和遺傳背景。常見病有:青霉素過敏、花粉病、支氣管哮喘、過敏性鼻炎、特應(yīng)性皮炎、食物過敏癥等。以哮喘為例,它是世界上最常見的慢性疾病之一。目前全世界有近2億患者,我國(guó)患病率接近1%,患病人數(shù)在1千萬(wàn)以。Ⅰ型超敏反應(yīng)主要由人免疫球蛋白E(human immunoglobulin E,IgE)介導(dǎo)發(fā)生,IgE是正常人血清中含量最少的Ig,血清濃度極低,約為5×10~(-5)mg/ml,主要由黏膜下淋巴結(jié)組織中的漿細(xì)胞分泌。IgE為親細(xì)胞抗體,其CH2和CH3結(jié)構(gòu)域可與肥大細(xì)胞、嗜堿性粒細(xì)胞上FC ε RI高親和力的結(jié)合,引起Ⅰ型超敏反應(yīng)�，F(xiàn)今抗IgE療法即用抗IgEmAb阻斷IgE或阻斷它的合成為包括哮喘在內(nèi)的過敏性疾病患者提供了極為有效的治療手段。但是,這些藥物存在有用藥量大、需長(zhǎng)期反復(fù)使用、易引起超敏反應(yīng)的缺陷。表位肽疫苗就是指用短的、合成的表位肽進(jìn)行免疫治療,只需少量注射就可以誘發(fā)產(chǎn)生抗體,可克服抗體及可溶性受體藥物的缺陷。因此IgE表位肽疫苗是治療Ⅰ型超敏反應(yīng)性疾病的一條有效途徑。噬菌體呈現(xiàn)肽庫(kù)是近年來發(fā)展很快的一項(xiàng)技術(shù),它已成為為已知分子尋找相應(yīng)配體的有效工具。因此本試驗(yàn)希望利用噬菌體篩選技術(shù)篩選得到IgE表位
[Abstract]:Type I hypersensitivity is also known as the rapid hypersensitivity, its main characteristics: rapid onset, physical functional disorder, obvious individual difference and genetic background. Common diseases are: penicillin allergy, hay fever, bronchial asthma, allergic rhinitis, atopic dermatitis, food allergy, and so on. As an example of asthma, it is one of the most common chronic diseases in the world. At present, there are nearly 200 million patients all over the world. The prevalence rate of China is close to 1%, and the number of diseases is 10 million. Type I hypersensitivity is mainly mediated by Human Immunoglobulin E (human immunoglobulin E, IgE). IgE is the least Ig in normal human serum, and its serum concentration is very low, which is about 5 10~ 10~ (-5) mg/ml, which is mainly secreted by plasma cells in submucosal lymph node tissue. IgE is a pro cell antibody, and its CH2 and CH3 domains can be combined with the high affinity of FC epsilon RI on mast cells and basophils, causing type I hypersensitivity. Anti - IgE therapy today is an extremely effective treatment for patients with allergic diseases, including asthma, using anti IgEmAb blocking IgE or blocking its synthesis. However, these drugs have a large amount of useful drugs, long-term repeated use, easy to cause hypersensitivity defects. Epitope peptide vaccine refers to immunotherapy with short and synthetic epitope peptides. Only a small amount of injection can induce antibodies, which can overcome the defects of antibodies and soluble receptor drugs. Therefore, the IgE epitope peptide vaccine is an effective way for the treatment of type I hypersensitivity disease. Phage display peptide library is a fast developing technique in recent years. It has become an effective tool for finding the corresponding ligands for known molecules. Therefore, we hope to use phage screening technique to screen the IgE epitopes
