【摘要】：目的篩選并鑒定埃及伊蚊(Aedes aegypti)全基因組中的yellow基因,分析其基因結(jié)構(gòu)、基因進(jìn)化以及在不同發(fā)育時(shí)期和不同組織的表達(dá)譜。方法運(yùn)用模式昆蟲(chóng)黑腹果蠅(Drosophila melanogaster)Dmyellow基因(Gen Bank登錄號(hào)為AAF45497)王漿主蛋白(MRJP)結(jié)構(gòu)域氨基酸序列,在埃及伊蚊全基因組數(shù)據(jù)庫(kù)中篩選并鑒定埃及伊蚊yellow基因家族,采用Ex PASy在線(xiàn)相關(guān)工具包分析其理化性質(zhì)和結(jié)構(gòu)域,運(yùn)用Signal P4.1預(yù)測(cè)信號(hào)肽,結(jié)合使用DNAstar、MAGA6.0和Gene Doc軟件包進(jìn)行同源性比對(duì)、保守性分析和系統(tǒng)進(jìn)化樹(shù)構(gòu)建。提取埃及伊蚊總RNA,合成c DNA,利用實(shí)時(shí)熒光定量PCR(q RT-PCR)法對(duì)埃及伊蚊不同發(fā)育時(shí)期(卵、Ⅰ~Ⅳ齡幼蟲(chóng)、蛹以及未吸血雌蚊和雄蚊)和未吸血雌蚊不同組織(唾液腺、中腸、脂肪體和卵巢)的yellow基因表達(dá)譜進(jìn)行相對(duì)定量分析。結(jié)果從埃及伊蚊全基因組中篩選出12條yellow基因(Aa-yellow、Aa-yellow-b、Aayellow-c、Aa-yellow-d、Aa-yellow-e、Aa-yellow-f2、Aa-yellow-fb、Aa-yellow-fc、Aa-yellow-g、Aa-yellow-g2、Aa-yellow-h和Aa-yellow-x)。生物信息學(xué)分析結(jié)果顯示,12條yellow基因均具有MRJP結(jié)構(gòu)域和信號(hào)肽序列。同源性比對(duì)結(jié)果顯示,埃及伊蚊yellow基因家族成員間同源性較低,為15%~49%;但其保守域間同源性較高,可達(dá)60%。系統(tǒng)進(jìn)化樹(shù)分析結(jié)果顯示,12個(gè)YELLOW蛋白分為5個(gè)亞家族,其中11個(gè)在黑腹果蠅中均有直系同源蛋白存在。q RT-PCR結(jié)果顯示,Aa-yellow-d和Aa-yellow-x在雄蚊中高表達(dá)(P0.01或P0.05),相對(duì)表達(dá)量分別為0.018 9和0.023 5;Aa-yellow-fc在雌蚊和唾液腺中特異高表達(dá)(P0.01),相對(duì)表達(dá)量分別為0.024 8和0.034 9;Aa-yellow-f2在Ⅱ齡幼蟲(chóng)中特異高表達(dá)(P0.01),相對(duì)表達(dá)量為0.093 4;Aa-yellow、Aa-yellow-e和Aa-yellow-fb在Ⅲ齡幼蟲(chóng)中特異高表達(dá)(P0.01),相對(duì)表達(dá)量分別為0.562 1、0.004 4和0.008 4;Aa-yellow、Aa-yellow-e、Aa-yellow-f2、Aayellow-fb、Aa-yellow-h和Aa-yellow-x等6條基因在卵巢中高表達(dá)(P0.01或P0.05),相對(duì)表達(dá)量分別為0.569 4、0.027 0、0.006 5、0.001 0、0.084 8和0.015 1;除Aa-yellow-c(0.004 0)和Aa-yellow-x(0.007 4)外,其余基因幾乎不在中腸表達(dá)。結(jié)論在埃及伊蚊基因組中獲得的12條yellow基因同源性較低,且在不同發(fā)育時(shí)期和不同組織中表達(dá)有差異。
