目的觀察皮膚再生醫(yī)療技術(shù)(moist exposed burn therapy/moist exposed burn ointment,MEBT/MEBO)對(duì)糖尿病大鼠創(chuàng)面組織中絲氨酸/蘇氨酸蛋白激酶-失調(diào)51樣激酶1(uncoordinated 51 like kinase-1,ULK1)表達(dá)的影響,探討MEBT/MEBO促進(jìn)糖尿病創(chuàng)面愈合的可能機(jī)制。方法選取SPF級(jí)Wistar雄性大鼠90只,隨機(jī)分為對(duì)照組、模型組和MEBO組,每組30只,其中對(duì)照組大鼠建立常規(guī)創(chuàng)面模型,模型組和MEBO組大鼠建立糖尿病創(chuàng)面模型。模型建立后,對(duì)照組及模型組大鼠創(chuàng)面予以生理鹽水紗布外敷治療,MEBO組大鼠創(chuàng)面予以濕潤(rùn)燒傷膏(moist exposed burn ointment,MEBO)藥紗外敷治療。分別于治療第1、11、18天采用Western blotting法檢測(cè)ULK1蛋白及qRT-PCR技術(shù)檢測(cè)ULK1 mRNA的表達(dá)水平,并對(duì)比觀察3組大鼠創(chuàng)面愈合情況。結(jié)果(1)治療第1天,3組大鼠創(chuàng)面均淤血水腫,無(wú)肉芽組織形成;治療第11天,對(duì)照組大鼠創(chuàng)面干燥,可見(jiàn)大片肉芽組織形成,模型組大鼠創(chuàng)面高度...
