構(gòu)建人多能性干細(xì)胞來源具有雙向分化潛能的肝臟類器官的研究

  我國(guó)是肝病大國(guó),各種原因?qū)е碌慕K末期肝病給人體健康帶來嚴(yán)重危害,給家庭和社會(huì)帶來沉重負(fù)擔(dān),也給護(hù)理人員帶來巨大壓力。隨著終末期肝病的發(fā)病率和死亡率逐年增加,護(hù)理人員在為越來越多的終末期肝病患者提供高質(zhì)量的護(hù)理方面發(fā)揮著至關(guān)重要的作用。終末期肝病目前公認(rèn)的唯一有效治療方式就是肝移植,但是肝源的匱乏明顯抑制了肝移植的廣泛應(yīng)用。近年來干細(xì)胞技術(shù)的快速發(fā)展給這一疾病的治療開辟了新的路徑,尤其是具有自我更新和向肝臟細(xì)胞和膽管細(xì)胞進(jìn)行雙向分化潛能的肝臟干細(xì)胞(HSCs)給肝病的治療提供了新的細(xì)胞來源。眾多的研究都在致力于如何獲取肝臟干細(xì)胞,目前獲取人肝臟干細(xì)胞的方式主要有兩種,一種是從人體肝臟組織中通過分離培養(yǎng)獲取原代肝臟干細(xì)胞,但是肝臟組織的獲得畢竟有限而且困難。另外一種重要的方式是從人多能性干細(xì)胞(hPSCs)包括胚胎干細(xì)胞(hESCs)和誘導(dǎo)多能性干細(xì)胞(hiPSCs)通過誘導(dǎo)分化獲得,由于hESCs已經(jīng)穩(wěn)定建系,hiPSCs可以取自病人皮膚細(xì)胞來建立,因此是HSCs的良好來源。但是目前由hPSCs誘導(dǎo)獲得HSCs需要依賴滋養(yǎng)細(xì)胞的支持、或者血清的添加、或者通過基因修飾的手段,這些操作方式復(fù)...
