小鼠脾臟、腹腔和骨髓源性巨噬細(xì)胞特征比較(英文)

目的:對(duì)比分析脾臟、腹腔和骨髓源性巨噬細(xì)胞(SPMs、PCMs和BMs)在安靜及極化狀態(tài)下的差異。創(chuàng)新點(diǎn):首次對(duì)比分析脾臟、腹腔和骨髓源性巨噬細(xì)胞在安靜(M0)及極化狀態(tài)(M1和M2)下的特征。方法:通過小鼠脾臟研磨及單細(xì)胞貼壁獲得脾源性巨噬細(xì)胞;腹腔灌洗及細(xì)胞貼壁獲得腹腔源性巨噬細(xì)胞;骨髓貼壁細(xì)胞在巨噬細(xì)胞集落刺激因子體外刺激下培養(yǎng)7天獲得骨髓源性巨噬細(xì)胞。三種細(xì)胞即為M0型巨噬細(xì)胞,M0在干擾素及脂多糖刺激下獲得M1型巨噬細(xì)胞,M0在白介素4(IL-4)刺激下獲得M2型巨噬細(xì)胞。通過流式細(xì)胞儀分析三種類型巨噬細(xì)胞在三種狀態(tài)下的Ⅱ類主要組織相容性復(fù)合體(MHC Ⅱ)和CD86表達(dá)差異。通過實(shí)時(shí)熒光定量聚合酶鏈?zhǔn)椒磻?yīng)(q PCR)檢測(cè)腫瘤壞死因子α(TNF-α)、白介素1β(IL-1β)、甘露糖受體(MR)和類幾丁質(zhì)酶3樣分子(Ym1)的表達(dá)變化。結(jié)論:骨髓貼壁細(xì)胞培養(yǎng)能獲得最大量的同源巨噬細(xì)胞(圖1和2),但表型偏向于M2型巨噬細(xì)胞(圖3和4)。三種巨噬細(xì)胞均能極化為M1和M2型巨噬細(xì)胞(圖5),其中SPMs具有更強(qiáng)的M1型極化能力,而M2型極化能力之間未見明顯差異(圖6)。綜上所述...
