MRTF-A調(diào)控miR-1273g-3p對Aβ 25-35 損傷的SH-SY5Y細胞自噬的影響及機制研究

目的:在Aβ25-35誘導人源性SH-SY5Y損傷細胞上,篩選能差異化表達并與MRTF-A相關的miRNA,探討MRTF-A介導miRNA對Aβ25-35誘導的SH-SY5Y細胞自噬的影響及機制。方法:1.在Aβ25-35誘導SH-SY5Y損傷細胞模型中,用基因芯片篩選與MRTF-A調(diào)控相關的miRNA:建立細胞模型;細胞分組(control,Aβ25-35,MRTF-A,MRTF-A+Aβ25-35)。轉(zhuǎn)染MRTF-A質(zhì)粒48h后,給予40μmol/L Aβ25-35處理,24h后,用western blot方法檢測MRTF-A的蛋白表達水平,并提取RNA。用Agilent RNA 6000 Nano/Pico Assay法檢測各組RNA質(zhì)量合格后,使用miRNA One Array芯片技術分析各組中發(fā)生顯著差異變化的miRNA。得到各組顯著變化的miRNA,再通過各組間相互比較得到MRTF-A調(diào)控的miRNA。篩選出差異性表達最為顯著的miRNA,并用...
