當(dāng)歸飲子對(duì)銀屑病模型豚鼠神經(jīng)酰胺含量、AQP-3基因及蛋白表達(dá)的干預(yù)研究

【摘要】:目的探討當(dāng)歸飲子對(duì)尋常型銀屑病模型豚鼠皮膚神經(jīng)酰胺(ceramide,Cer)含量、水通道蛋白3(aquaporin 3,AQP-3)基因及蛋白表達(dá)的干預(yù)影響。方法建立尋常型銀屑病動(dòng)物模型,分為當(dāng)歸飲子組、甲氨蝶呤組、生理鹽水組、模型對(duì)照組、空白對(duì)照組,每組8只,酶聯(lián)免疫吸附實(shí)驗(yàn)(ELISA)法檢測(cè)造模皮損部位Cer含量,逆轉(zhuǎn)錄PCR(RT-PCR)和免疫印跡試驗(yàn)(Western blot)法檢測(cè)造模皮損部位AQP-3基因及蛋白表達(dá),分析尋常型銀屑病動(dòng)物模型皮損與皮膚屏障功能密切相關(guān)的基因及蛋白表達(dá)的變化。結(jié)果豚鼠耳部皮膚銀屑病模型造模成功。模型對(duì)照組皮損中ELISA法檢測(cè)Cer含量,以及RT-PCR和Western blot法檢測(cè)AQP-3基因的蛋白表達(dá)均較低;當(dāng)歸飲子組與甲氨蝶呤組和各組中Cer含量、AQP-3基因蛋白表達(dá)增強(qiáng)(P0.01),且當(dāng)歸飲子組較甲氨蝶呤組增強(qiáng)明顯(P0.01)。結(jié)論當(dāng)歸飲子能增強(qiáng)銀屑病模型豚鼠皮膚Cer含量及AQP-3基因和蛋白表達(dá),對(duì)改善銀屑病動(dòng)物模型的皮膚屏障功能作用確切,效果優(yōu)于甲氨蝶呤組。
[Abstract]:Objective to investigate the effect of Angelica sinensis decoction on the content of ceramide (ceramide,Cer) and the expression of aquaporin 3 (aquaporin 3) gene and protein in guinea pigs with psoriasis vulgaris. Methods the animal model of psoriasis vulgaris was established and divided into Angelica sinensis group, methotrexate group, saline group, model control group and blank control group with 8 rats in each group. The content of Cer in the lesion site was detected by enzyme-linked immunosorbent assay (ELISA), and the expression of AQP-3 gene and protein in the lesion site was detected by reverse transcription PCR (RT-PCR) and immunoblotting (Western blot). To analyze the changes of gene and protein expression in psoriatic vulgaris model, which are closely related to skin barrier function. Results the model of psoriasis in the ear of guinea pigs was successfully established. The content of Cer in the skin lesions of the model control group was detected by ELISA, and the protein expression of AQP-3 gene was detected by RT-PCR and Western blot. The content of Cer and the expression of AQP-3 gene protein in Angelica sinensis group and methotrexate group and each group were increased (P 0.01), and the expression of AQP-3 gene protein in Angelica sinensis group was significantly higher than that in methotrexate group (P 0.01). Conclusion Angelica sinensis decoction can enhance the content of Cer and the expression of AQP-3 gene and protein in the skin of psoriatic guinea pigs, and has a definite effect on improving the skin barrier function of psoriatic animal model, and the effect is better than that of methotrexate group.
