種子育麟散對超促排卵模型大鼠子宮內(nèi)膜整合素αvβ3、HOXA10表達(dá)的影響

【摘要】:目的 探討種子育麟散治療不孕癥的可能作用機(jī)制。方法 選取30只SD雌性大鼠隨機(jī)分為空白對照組、模型組、中藥組,每組10只。模型組和中藥組通過腹腔內(nèi)注射丙氨瑞林、尿促性腺激素、注射用絨毛膜促性腺激素建立控制性超促排卵模型大鼠。造模開始的同時(shí)中藥組給予種子育麟散7.2 g/(kg·d)灌胃,模型組、空白對照組給予等量生理鹽水灌胃,均每日1次,每次2 ml,連續(xù)9天。于孕5天后分別采用免疫組化法、Western blot法檢測大鼠子宮內(nèi)膜整合素αvβ3、同源框(HOX)A10蛋白表達(dá),RT-PCR法檢測HOXA10、整合素αvβ3 mRNA表達(dá)。結(jié)果 與空白對照組比較,模型組子宮內(nèi)膜整合素αvβ3、HOXA10蛋白及HOXA10、整合素αvβ3 mRNA表達(dá)量明顯減少(P0.05);與模型組比較,中藥組子宮內(nèi)膜整合素αvβ3、HOXA10蛋白及整合素αvβ3、HOXA10 mRNA表達(dá)量明顯增加(P0.05)。結(jié)論 種子育麟散能夠提高大鼠子宮內(nèi)膜整合素αvβ3、HOXA10蛋白及mRNA的表達(dá),從而改善妊娠結(jié)局,可能是其治療不孕癥的機(jī)制之一。
[Abstract]:Objective to explore the possible mechanism of seed Yulin Powder in the treatment of sterility. Methods 30 female SD rats were randomly divided into blank control group (n = 10), model group (n = 10) and traditional Chinese medicine group (n = 10). In the model group and the traditional Chinese medicine group, the controlled superovulatory model rats were established by intraabdominal injection of propyrine, urinary gonadotropin and chorionic gonadotropin. At the beginning of the model, the traditional Chinese medicine group was given seed Yulin Powder 7.2g / (kg 路d) intragastrically, and the model group and the blank control group were given the same amount of saline once a day for 2 ml, for 9 days. After 5 days of pregnancy, the expression of integrin 偽 v 尾 3 and homeobox (HOX) A10 protein in rat endometrium was detected by, Western blot method, and the expression of HOXA10, integrin 偽 v 尾 3 mRNA was detected by RT-PCR assay. Results compared with the blank control group, the expression of integrin 偽 v 尾 3, HoxA10 protein and HOXA10, integrin 偽 v 尾 3 mRNA in the model group was significantly lower than that in the model group (P 0.05). Compared with the model group, the expression of integrin 偽 v 尾 3, HOXA10 protein and integrin 偽 v 尾 3, HOXA10 mRNA increased significantly in the model group (P 0.05). Conclusion seed Yulin Powder can increase the expression of integrin 偽 v 尾 3, HoxA10 protein and mRNA in rat endometrium, thus improving the pregnancy outcome, which may be one of the mechanisms of its treatment of sterility.
