【摘要】：【目的】利用光鑷?yán)庾V技術(shù)(Laser Tweezers Raman Spectroscopy,LTRS)研究金黃色葡萄球菌(Staphylococcus aureus)細(xì)胞內(nèi)主要生物大分子含量的動態(tài)變化,探尋監(jiān)測微生物代謝狀態(tài)變化的有效工具。【方法】收集分批培養(yǎng)過程不同時間點(diǎn)金黃色葡萄球菌細(xì)胞的拉曼光譜,觀察其特征峰依賴時間的變化特點(diǎn)。【結(jié)果】歸屬于核酸的特征峰,如667cm~(-1)峰,724cm~(-1)峰,781cm~(-1)峰,809cm~(-1)峰和1 574cm~(-1)峰在整個分批生長階段持續(xù)降低,在對數(shù)生長期下降幅度尤其劇烈,反映了對數(shù)生長期細(xì)胞分裂快速,DNA復(fù)制活躍;與蛋白質(zhì)相關(guān)的特征峰,如850cm~(-1)峰,1 001cm~(-1)峰和1 662cm~(-1)峰,在對數(shù)生長期顯著增強(qiáng),而到穩(wěn)定期峰強(qiáng)度基本維持恒定。【結(jié)論】光鑷?yán)庾V技術(shù)能實(shí)時監(jiān)測生物樣品內(nèi)生物大分子的含量,是單細(xì)胞水平上研究微生物代謝狀態(tài)變化的有效工具。
[Abstract]:[objective] to study the dynamic changes of main biological macromolecules in Staphylococcus aureus (Staphylococcus aureus) cells by optical tweezers Raman spectroscopy (Laser Tweezers Raman Spectroscopy,LTRS), and to explore an effective tool to monitor the changes of microbial metabolism. [methods] Raman spectra of Staphylococcus aureus cells at different time points during batch culture were collected. The variation of characteristic peak dependent time was observed. [results] the characteristic peaks attributed to nucleic acid, such as 667cm1 peak, 724cm1 peak, 781cm1 peak, 809cm1 peak and 1574 cm1 peak, decreased continuously in the whole batch growth stage, especially in logarithmic growth period, which reflected the rapid cell division and active DNA replication in logarithmic growth period. The characteristic peaks related to protein, such as 850 cm ~ (- 1) peak, 1 001 cm ~ (- 1) peak and 1 662 cm ~ (- 1) peak, increased significantly during logarithmic growth period, but remained constant at stable stage. [conclusion] Optical tweezers Raman spectroscopy can monitor the content of biological macromolecules in biological samples in real time, and it is an effective tool to study the changes of microbial metabolism at single cell level.
