【摘要】：目的:觀察柴貝止癇湯、卡馬西平、柴貝止癇湯與卡馬西平聯(lián)用3種不同干預(yù)對難治性癲癇大鼠模型行為學(xué)及P-糖蛋白(P-gp)和MDR1表達(dá)的影響。方法:采用側(cè)腦室注射海人酸法制作癲癇持續(xù)狀態(tài)后慢性難治性癲癇大鼠模型,分為模型組、西藥組、中藥組、中西藥組、正常組、假手術(shù)組6組。給藥時間3個月。以Western Blot及實(shí)時熒光定量PCR法檢測治療1、3個月海馬P-gp及MDR1a/1b mRNA水平。結(jié)果:海馬中P-gp、MDR1a/1b mRNA含量,模型組高于假手術(shù)組,在治療1個月差異具有統(tǒng)計學(xué)意義(P0.01,P0.05);西藥組高于模型組,差異不具統(tǒng)計學(xué)意義;中藥組、中西藥組低于模型組,其中P-gp含量在治療1月時差異具有統(tǒng)計學(xué)意義(P0.05);中西藥組低于西藥組,差異具有統(tǒng)計學(xué)意義(P0.05)。結(jié)論:柴貝止癇湯能夠抑制難治性癲癇P-gp表達(dá),這可能通過對MDR1a/1b mRNA轉(zhuǎn)錄水平的影響實(shí)現(xiàn)。
[Abstract]:Aim: to observe the effects of Chaibei Zhixian decoction, carbamazepine, Chaibei Zhixian decoction and carbamazepine on behavior and expression of P-glycoprotein (P-gp) and MDR1 in refractory epileptic rats. Methods: the rat model of chronic refractory seizures after persistent status was established by intracerebroventricular injection of kainic acid. The rats were divided into 6 groups: model group, western medicine group, Chinese and western medicine group, normal group and false operation group. The drug was given for 3 months.
