高溫致鼠神經(jīng)管畸形神經(jīng)上皮中N-cadherin、bFGF、PCNA的表達(dá)及其意義研究

【摘要】:目的:研究高溫對(duì)鼠胚神經(jīng)管發(fā)育時(shí)期神經(jīng)上皮細(xì)胞的粘附聚集、增殖凋亡及細(xì)胞生長(zhǎng)因子的影響,從而探討高溫致神經(jīng)管畸形的發(fā)生機(jī)理。方法:將孕鼠隨機(jī)分成為實(shí)驗(yàn)組30只和對(duì)照組25只,實(shí)驗(yàn)組鼠于孕8d下午15:30進(jìn)行42℃水浴20min,對(duì)照組鼠置于37℃水浴箱20min。實(shí)驗(yàn)組和對(duì)照組各10只孕鼠于孕15d處死剖腹取胎,辨認(rèn)和計(jì)數(shù)活胎、死胎、吸收胎和畸胎。其余孕鼠35只,分別于37℃和42°C水浴后2h、8h、16h、24h、48h處死,剖腹取胎,置于10%的中性福爾馬林液中固定,常規(guī)石蠟包埋、連續(xù)切片。用免疫組織化學(xué)法和圖像分析儀檢測(cè)N-cadherin、bFGF、PCNA在各期實(shí)驗(yàn)組和對(duì)照組鼠胚神經(jīng)管上皮細(xì)胞的表達(dá)變化。結(jié)果:①實(shí)驗(yàn)組鼠胚神經(jīng)管畸形率達(dá)70.91%,對(duì)照組致畸率為0。②N-cadherin的免疫組織化學(xué)結(jié)果:水浴后2-8h N-cadherin在對(duì)照組及實(shí)驗(yàn)組神經(jīng)管上皮細(xì)胞表達(dá)呈陽性,二者之間的OD值差異無統(tǒng)計(jì)學(xué)意義,內(nèi)外界膜、脊索及神經(jīng)管周圍間充質(zhì)細(xì)胞也可見陽性反應(yīng)。水浴后16-24h, N-cadherin在對(duì)照組神經(jīng)管上皮細(xì)胞表達(dá)呈強(qiáng)陽性,而在實(shí)驗(yàn)組表達(dá)呈弱陽性,OD值差異有統(tǒng)計(jì)學(xué)意義(p0.01),周圍組織陽性變化不明顯。至水浴后48h實(shí)驗(yàn)組與對(duì)照組相比,神經(jīng)上皮N-cadherin免疫陽性反應(yīng)差異不明顯,N-cadherin蛋白在兩組神經(jīng)管組織表達(dá)均減弱。③)bFGF的免疫組織化學(xué)結(jié)果:水浴后2h、8h、16h bFGF蛋白在實(shí)驗(yàn)組和對(duì)照組神經(jīng)上皮中表達(dá),兩組OD值差異無統(tǒng)計(jì)學(xué)意義,水浴后24h-48h實(shí)驗(yàn)組神經(jīng)上皮的表達(dá)弱于對(duì)照組,且OD值差異有統(tǒng)計(jì)學(xué)意義(p0.05), bFGF在兩組神經(jīng)管上皮的表達(dá)隨胚齡增加有增強(qiáng)的趨勢(shì),內(nèi)外界膜、脊索和周圍間充質(zhì)也呈現(xiàn)陽性反應(yīng)。④孕8d, PCNA蛋白廣泛表達(dá)在神經(jīng)管組織,神經(jīng)管上皮細(xì)胞、神經(jīng)嵴細(xì)胞、周圍間充質(zhì)細(xì)胞均呈現(xiàn)高表達(dá)狀態(tài)。水浴后2h和8h, PCNA在實(shí)驗(yàn)組神經(jīng)組織的陽性表達(dá)數(shù)低于對(duì)照組,但差異無統(tǒng)計(jì)學(xué)意義,高溫處理后16h、24h和48h PCNA在實(shí)驗(yàn)組神經(jīng)管上皮組織的陽性細(xì)胞數(shù)低于對(duì)照組,且差異有統(tǒng)計(jì)學(xué)意義(p0.05)。水浴處理24h之后,PCNA在實(shí)驗(yàn)組和對(duì)照組神經(jīng)組織中的陽性細(xì)胞數(shù)均有明顯下降的趨勢(shì)。結(jié)論:高溫可使N-cadherin、bFGF、PCNA在神經(jīng)管上皮細(xì)胞的表達(dá)降低,影響細(xì)胞間的識(shí)別、粘附,干擾細(xì)胞增殖與凋亡的動(dòng)態(tài)平衡,誘導(dǎo)NTDs發(fā)生,是高溫導(dǎo)致神經(jīng)管缺陷的重要途徑之一。
