靶向哺乳動(dòng)物細(xì)胞線粒體的核酸轉(zhuǎn)運(yùn)

【摘要】:線粒體DNA(mitochondrial DNA,mtDNA)的遺傳性突變和缺陷是多種線粒體功能失調(diào)相關(guān)疾病的根本原因。靶向線粒體遞送核酸,可從根本上糾正mtDNA突變、挽救mtDNA損傷、阻斷疾病進(jìn)程。哺乳細(xì)胞內(nèi)線粒體的核酸轉(zhuǎn)運(yùn)途徑與細(xì)胞核的基因轉(zhuǎn)染大不相同。該文綜述了向哺乳動(dòng)物細(xì)胞線粒體遞送DNA和RNA(tRNA、rRNA、mRNA和反義RNA)的有效策略,并對(duì)其存在問(wèn)題和發(fā)展趨勢(shì)做一闡述。
[Abstract]:The genetic mutation and defect of mtDNA (DNA (mitochondrial DNA,mtDNA) are the root causes of many diseases related to mtDNA dysfunction. The delivery of nucleic acid to mitochondria can fundamentally correct mtDNA mutation, save mtDNA damage and block the process of disease. The nucleic acid transport pathway of mitochondria in lactation cells is very different from that of nuclear gene transfer. In this paper, the effective strategies of delivering DNA and RNA (tRNA,rRNA,mRNA and antisense RNA) to mammal cells are reviewed, and their existing problems and development trends are described.
