血清和糖皮質(zhì)激素調(diào)節(jié)蛋白激酶1在CNS疾病中的作用研究新進(jìn)展

【摘要】:血清和糖皮質(zhì)激素調(diào)節(jié)蛋白激酶1(SGK1)是1993年在受糖皮質(zhì)激素刺激的大鼠乳腺癌細(xì)胞中發(fā)現(xiàn)的一個絲氨酸/蘇氨酸蛋白激酶。與多數(shù)蛋白激酶不同的是,血清和糖皮質(zhì)激素調(diào)節(jié)蛋白激酶1不僅受翻譯后的磷酸化/去磷酸化調(diào)節(jié),而且存在轉(zhuǎn)錄水平上的快速調(diào)節(jié)機(jī)制。它具有廣泛的生理功能,如調(diào)節(jié)離子通道,介導(dǎo)細(xì)胞增殖、分化、存活及凋亡的信號轉(zhuǎn)導(dǎo)等。它與帕金森病、阿爾茨海默病等多種中樞神經(jīng)系統(tǒng)疾病密切相關(guān),在神經(jīng)元變性、凋亡過程中起著重要作用。本文將從血清和糖皮質(zhì)激素調(diào)節(jié)蛋白激酶1的結(jié)構(gòu)、生理功能、與中樞神經(jīng)系統(tǒng)疾病的關(guān)系及其抑制劑等方面的新進(jìn)展作一綜述。
[Abstract]:Serum and glucocorticoid regulated protein kinase 1 (SGK1) is a serine / threonine protein kinase found in rat breast cancer cells stimulated by glucocorticoid in 1993. Unlike most protein kinases, serum and glucocorticoid regulated protein kinase 1 is not only regulated by post-translation phosphorylation / dephosphorylation, but also by a rapid regulatory mechanism at the transcriptional level. It has a wide range of physiological functions, such as regulating ion channels, mediating cell proliferation, differentiation, survival and apoptosis signal transduction and so on. It is closely related to Parkinson's disease, Alzheimer's disease and other central nervous system diseases, and plays an important role in neuronal degeneration and apoptosis. In this paper, the structure and physiological function of serum and glucocorticoid regulated protein kinase 1, the relationship between serum and glucocorticoid regulated protein kinase 1 and central nervous system diseases and their inhibitors are reviewed.
