開結(jié)湯對腺嘌呤所致慢性腎衰竭模型大鼠TGF-β1、MCP-1表達的影響

【摘要】:目的研究開結(jié)湯對腺嘌呤所致慢性腎衰竭模型大鼠TGF-β1、MCP-1表達的影響。方法選取60只SD大鼠按體質(zhì)量隨機分為6組:空白對照組、模型組、開結(jié)湯低、中、高劑量治療組、尿毒清治療組,除空白組外,其余5組連續(xù)灌服腺嘌呤21 d以復制慢性腎衰竭大鼠模型,劑量均是300 mg/(kg·d)。造模成功后用開結(jié)湯、尿毒清進行干預,治療30 d后,心臟取血檢測常規(guī)生化指標,取大鼠腎組織,用免疫組化方法檢測TGF-β1、MCP-1的表達。結(jié)果與正常組比較,模型組大鼠血清中BUN、Scr、P的含量及腎組織中TGF-β1、MCP-1的表達顯著增高,差異有統(tǒng)計學意義(P0.05);經(jīng)過藥物治療,開結(jié)湯3個劑量組及尿毒清組大鼠血清中BUN、Scr、P的含量及腎組織中TGF-β1、MCP-1的表達明顯低于模型組(P0.05);開結(jié)湯3個劑量組與尿毒清組比較,低、中劑量組在降低血清BUN、Scr、P的含量及抑制TGF-β1、MCP-1在腎組織中表達方面較尿毒清組顯著(P0.05)。結(jié)論開結(jié)湯可以改善腎功能,保護腎臟。作用途徑是抑制TGF-β1及MCP-1在腎組織中的表達,減少炎性細胞浸潤,從而延緩腎纖維化進程。
[Abstract]:Objective to study the effect of Kaijie decoction on the expression of TGF- 尾 1 and MCP-1 in rats with chronic renal failure induced by adenine. Methods 60 SD rats were randomly divided into 6 groups according to body mass: blank control group, model group, low, middle and high dose Kaijie decoction treatment group, Niaoduqing treatment group, except blank group, The other five groups were given adenine for 21 days to establish the rat model of chronic renal failure at a dose of 300 mg/ (kg 路d). After successful establishment of the model, Kaijie decoction and Niaoduqing were used to intervene. After 30 days of treatment, the routine biochemical indexes were detected by heart blood collection, the kidney tissue of rats was taken, and the expression of TGF- 尾 1 and MCP-1 was detected by immunohistochemical method. Results compared with the normal group, the content of BUN,Scr,P in serum and the expression of TGF- 尾 1 and MCP-1 in renal tissue in the model group were significantly higher than those in the normal group (P 0.05). After drug treatment, the content of BUN,Scr,P in serum and the expression of TGF- 尾 1 and MCP-1 in renal tissue of rats in three dose groups of Kaijie decoction and Niaoduqing group were significantly lower than those in model group (P 0.05). Compared with Niaoduqing group, Kaijie decoction three dose groups were lower than Niaoduqing group in decreasing the content of serum BUN,Scr,P and inhibiting the expression of TGF- 尾 1 and MCP-1 in renal tissue (P 0.05). Conclusion Kaijie decoction can improve renal function and protect kidney. The pathway is to inhibit the expression of TGF- 尾 1 and MCP-1 in renal tissue and reduce the infiltration of inflammatory cells, thus delaying the process of renal fibrosis.
