肺炎球菌多糖衍生物中吡啶硼烷殘留量核磁共振檢測方法的建立

【摘要】:目的:建立核磁共振(nuclear magnetic resonance,NMR)方法檢測肺炎球菌多糖衍生物中吡啶硼烷殘留量的方法。方法:建立1H-NMR檢測方法:脈沖程序?yàn)樗鍓褐?溫度為298 K(25℃)。具體參數(shù)設(shè)置如下:中心頻率約為4.7 ppm左右;圖譜寬度為7 211 Hz;采樣時(shí)間為2.272 s;弛豫延遲時(shí)間為2 s;掃描次數(shù)為512次;信號經(jīng)傅里葉轉(zhuǎn)換后加入1.0 Hz的窗口函數(shù),檢測結(jié)果使用Topspin 3.0軟件進(jìn)行處理。并驗(yàn)證該方法的專屬性、檢測限及耐用性。結(jié)果:該方法檢測結(jié)果顯示溶劑對所測組分無干擾;吡啶硼烷與肺炎球菌多糖衍生物有各自明顯的特征峰,兩者混合之后信號無交叉重疊,并都可以明顯檢測到各自的信號,說明方法專屬性良好。吡啶硼烷濃度在1%~0.01%范圍內(nèi)均可以明顯檢測到Py-BH3信號,且S/N良好。最終最低檢測限定為0.02%。改變試驗(yàn)溫度化學(xué)位移及信噪比未發(fā)生明顯變化,方法顯示耐用性良好。結(jié)論:建立的1H-NMR檢測方法檢測肺炎球菌多糖衍生物中吡啶硼烷殘留具有極高的專屬性和耐用性,可用于肺炎球菌結(jié)合疫苗中吡啶硼烷殘留量的測定。
