愈癲湯對地卓西平所致精神分裂癥模型大鼠氧化應(yīng)激防御系統(tǒng)的影響

【摘要】:目的:基于多巴胺(DA)神經(jīng)元功能亢進(jìn)理論,觀察愈癲湯對精神分裂癥模型大鼠額前腦組織抗氧化應(yīng)激防御系統(tǒng)相關(guān)指標(biāo)的影響。方法:將48只雄性成年SD大鼠隨機(jī)分為空白組、模型組、對照組及治療組,每組12只。其中模型組、對照組及治療組應(yīng)用地卓西平腹腔注射造模,空白組應(yīng)用0.9%氯化鈉溶液行相同方法腹腔注射,連續(xù)14d;于第15天進(jìn)行干預(yù)性給藥,對照組給予0.036mg/m L利培酮懸濁液灌胃,治療組給予0.92g/m L愈癲湯水煎劑灌胃,空白組、模型組給予相應(yīng)體積蒸餾水灌胃,連續(xù)14d;檢測各組大鼠額前腦組織超氧化物歧化酶(SOD)、谷胱甘肽過氧化物酶(GSH)、過氧化氫酶(CAT)活性及DA的表達(dá)情況。結(jié)果:治療組大鼠額前組織內(nèi)SOD、GSH及CAT活性明顯高于模型組(P0.05),而DA的表達(dá)明顯低于模型組(P0.05)。結(jié)論:愈癲湯可通過促進(jìn)腦組織中SOD、GSH及CAT活性,將超氧化物轉(zhuǎn)化成無毒的水,從而抑制自由基的形成,阻斷氧化應(yīng)激反應(yīng),從而逆轉(zhuǎn)DA神經(jīng)元功能亢進(jìn)引起的氧化應(yīng)激反應(yīng)。
[Abstract]:Aim: based on the theory of dopaminergic (DA) neuron hyperfunction, the effects of Yuwei decoction on the indexes of antioxidant stress defense system in frontal forebrain of schizophrenic rats were observed. Methods: 48 adult male SD rats were randomly divided into blank group, model group, control group and treatment group with 12 rats in each group. The model group, control group and treatment group were injected intraperitoneally with diazepine, and the blank group were injected intraperitoneally with 0.9% sodium chloride solution for 14 days. On the 15th day, the control group was given 0.036mg/m-L risperidone suspension by gastric perfusion, the treatment group was given 0.92g/m-L Yuyang decoction, the blank group, and the model group were given corresponding volume distilled water for 14 days. The activity of superoxide dismutase (SOD), glutathione peroxidase (GSH), catalase (CAT) (CAT) and the expression of DA in prefrontal brain tissue of rats were measured. Results: the activities of SOD,GSH and CAT in prefrontal tissue in the treatment group were significantly higher than those in the model group (P0.05), while the expression of DA in the treatment group was significantly lower than that in the model group (P0.05). Conclusion: Yuwei decoction can inhibit the formation of free radicals and block the oxidative stress reaction by promoting the activity of SOD,GSH and CAT in brain tissue and transforming superoxide into non-toxic water. Thus reverse the oxidative stress response induced by hyperfunction of DA neurons.
