【摘要】：目的研究大鼠發(fā)育過(guò)程中,生長(zhǎng)休止蛋白7(growth arrest-specific protein 7,Gas7)在斜角帶核(diagonal band nucleus,DB)的表達(dá)。方法應(yīng)用RT-PCR方法、Western blot方法和免疫組織化學(xué)方法觀(guān)察胚胎期16.5天(E16.5)、E20.5、出生當(dāng)天(P0)、生后7天(P7)、P14、P21和2月齡(成年)大鼠DB的Gas7表達(dá)及定位。結(jié)果 RT-PCR和Western blot檢測(cè)顯示,在E16.5時(shí)Gas7表達(dá)最弱,其后逐漸增強(qiáng),至P21時(shí)達(dá)到高峰;免疫組織化學(xué)染色顯示,斜角帶核水平支(horizontal limb of the diagonal band,HDB)在E16.5和E20.5時(shí)出現(xiàn)Gas7免疫反應(yīng)陽(yáng)性產(chǎn)物,Gas7陽(yáng)性神經(jīng)元出現(xiàn)于P0,至P21時(shí)陽(yáng)性神經(jīng)元數(shù)量最多,染色最深;斜角帶核垂直支(vertical limb ofthe diagonal band,VDB)至P14時(shí)Gas7陽(yáng)性神經(jīng)元最多,P21和成年均有所減少。結(jié)論 Gas7的表達(dá)在大鼠DB發(fā)育過(guò)程中具有時(shí)間和空間上的特異性,提示Gas7可能參與大鼠DB的發(fā)育過(guò)程。
[Abstract]:Aim to study the expression of growth rest protein 7 (growth arrest-specific protein 7, Gas 7) in diagonal band nucleus (diagonal band nucleus,DB) during development in rats. Methods RT-PCR, Western blot method and immunohistochemical method were used to observe the embryonic stage of 16.5 days (E16.5), E20.5, the day of birth (P0), postnatal day 7 (P7), P14, Expression and localization of Gas7 in DB of P21 and 2 month old rats. Results RT-PCR and Western blot showed that the expression of Gas7 was the weakest at E16.5, then increased gradually and reached the peak at P21. Immunohistochemical staining showed that (horizontal limb of the diagonal band,HDB in the horizontal branch of the diagonal band showed Gas7 immunoreactive products at E16.5 and E20.5, Gas7 positive neurons appeared in P0, and the number of positive neurons at P21 was the most, and the number of positive neurons at E16.5 and E20.5 was more than that at E16.5 and E20.5, respectively. The staining was the deepest; From the vertical branch of the diagonal band nucleus (vertical limb ofthe diagonal band,VDB) to P14, the number of Gas7-positive neurons was the most, while the number of P21 and adult neurons was decreased. Conclusion the expression of Gas7 has temporal and spatial specificity during the development of rat DB, suggesting that Gas7 may be involved in the development of DB in rats.
