天麻素對不同生理病理狀態(tài)下CINP大鼠模型的作用

【摘要】:目的研究天麻素(Gastrodin)對腫瘤、非腫瘤2種不同生理病理狀態(tài)下化療誘導(dǎo)的神經(jīng)病理性疼痛(化療痛,Chemotherapy-induced Neuropathic Pain,CINP)大鼠模型的作用。方法采用腹腔注射長春新堿(Vincristine,VCR,125μg·kg~(-1))建立正常大鼠和Walker-256乳腺癌荷瘤大鼠CINP動物模型,并腹腔注射給予不同劑量的天麻素(60,120 mg·kg~(-1));采用Von Frey電子測痛儀檢測大鼠機械刺激痛閾值,PL-200型熱刺痛儀測定大鼠熱刺激痛閾值。結(jié)果 VCR對腫瘤、非腫瘤大鼠均可誘導(dǎo)產(chǎn)生機械痛敏反應(yīng)和熱痛敏反應(yīng),使其痛閾值均明顯降低(P0.01);天麻素可劑量依賴性抑制腫瘤、非腫瘤CINP大鼠模型的機械痛敏反應(yīng)和熱痛敏反應(yīng)(P0.01)。將大鼠注射VCR前后的痛閾值差值以及天麻素治療前后大鼠痛閾值差值進一步比較,VCR對非腫瘤組大鼠機械痛痛閾值的降低幅度比腫瘤組的明顯(P0.01),而VCR對腫瘤組的熱痛閾值降低幅度比非腫瘤組明顯,但差異無統(tǒng)計學(xué)意義(P0.05);天麻素對非腫瘤組大鼠機械痛閾值的提高作用比腫瘤組明顯(P0.05),而天麻素對腫瘤組大鼠的熱痛閾值提高作用比非腫瘤組明顯(P0.05)。結(jié)論天麻素對VCR誘導(dǎo)的腫瘤、非腫瘤大鼠CINP均有劑量依賴性抑制作用;但對生理、病理狀態(tài)下大鼠的不同感覺神經(jīng)纖維的痛覺抑制作用敏感性存在差異,對VCR誘導(dǎo)痛敏反應(yīng)越敏感的神經(jīng)纖維其抑制作用也相對越強。
[Abstract]:Aim to study the effect of gastrodin (Gastrodin) on the neuropathic pain (Chemotherapy-induced Neuropathic Pain,CINP) model induced by chemotherapy in two different physiological and pathological states of tumor and non-tumor. Methods CINP models of normal rats and Walker-256 rats bearing breast cancer were established by intraperitoneal injection of vincristine (Vincristine,VCR, 125 渭 g kg~ (- 1), and different doses of gastrodin (60120 mg kg~ (- 1);) were injected intraperitoneally. The pain threshold of mechanical stimulation in rats was measured by Von Frey electronic pain meter, and the pain threshold of thermal stimulation in rats was measured by PL- 200 hot prickly pain meter. Results VCR could induce mechanical hyperalgesia and thermal hyperalgesia in both tumor and non-tumor rats, resulting in a significant decrease in pain threshold (P0.01). Gastrodin could inhibit the mechanical hyperalgesia and thermal hyperalgesia in tumor-free and non-tumor CINP rats in a dose-dependent manner (P0.01). The difference of pain threshold before and after injection of VCR and the difference of pain threshold before and after gastrodin treatment in rats were further compared. The decrease of mechanical pain threshold in non-tumor group was significantly lower than that in tumor group (P0.01), and the decrease in mechanical pain threshold in non-tumor group was more obvious than that in tumor group (P0.01). However, the decrease of thermal pain threshold in VCR group was more obvious than that in non-tumor group, but there was no significant difference (P0.05). The effect of gastrodin on mechanical pain threshold of non-tumor group was significantly higher than that of tumor group (P0.05), while the effect of gastrodin on thermal pain threshold of non-tumor group was significantly higher than that of non-tumor group (P0.05). Conclusion Gastrodin has a dose-dependent inhibitory effect on VCR-induced tumor and non-tumor rat CINP in a dose-dependent manner. However, there were differences in sensitivity to pain inhibition of different sensory nerve fibers under physiological and pathological conditions. The more sensitive nerve fibers were to VCR-induced hyperalgesia, the stronger their inhibitory effects were.
