銀杏葉黃酮對(duì)急性腦梗死模型小鼠凋亡因子的影響

【摘要】:目的研究銀杏葉黃酮對(duì)急性腦梗死模型小鼠血清低氧誘導(dǎo)因子-1α(HIF-1α)、腫瘤壞死因子-α(TNF-α)及半胱天冬酶-3(caspase-3)影響。方法選取昆明雌性小鼠24只,隨機(jī)選取8只為假手術(shù)組,剩下小鼠進(jìn)行急性腦梗死模型的構(gòu)建。將模型小鼠按照體重隨機(jī)分成模型組和實(shí)驗(yàn)組,每組8只,實(shí)驗(yàn)組每天腹腔注射20 mg·kg~(-1)銀杏葉黃酮,假手術(shù)組和模型組同樣給藥方式給予0.9%NaCl,連續(xù)7 d。處死小鼠,用酶聯(lián)免疫吸附法檢測(cè)各組急性腦梗死模型小鼠血清中HIF-1α、TNF-α及Caspase-3的表達(dá)水平。結(jié)果假手術(shù)組、模型組、實(shí)驗(yàn)組小鼠血清中的TNF-α分別為(5.16±1.04),(33.02±20.34),(22.64±10.17)ng·mL~(-1);這3組小鼠血清中的HIF-1α分別為(508.94±123.18),(735.19±324.05),(626.24±212.76)ng·mL~(-1);這3組小鼠血清中的Caspase-3分別為(6.54±0.24),(12.23±3.45),(10.24±1.58)ng·mL~(-1);模型組小鼠血清中TNF-α、HIF-1α、Caspase-3表達(dá)明顯高于假手術(shù)組,差異均有統(tǒng)計(jì)學(xué)意義(均P0.05);實(shí)驗(yàn)組小鼠血清中TNF-α、HIF-1α、Caspase-3表達(dá)明顯低于模型組,差異均有統(tǒng)計(jì)學(xué)意義(均P0.05)。結(jié)論銀杏葉黃酮能夠明顯下調(diào)急性腦梗死小鼠血清中TNF-α、Caspase-3及HIF-1α水平。
[Abstract]:Objective to study the effects of ginkgo biloba flavonoids on serum hypoxia inducible factor-1 偽 (HIF-1 偽), tumor necrosis factor- 偽 (TNF- 偽) and cysteinase 3 (caspase-3) in mice with acute cerebral infarction. Methods Twenty-four Kunming female mice were selected and 8 mice were randomly selected as sham-operated group. The remaining mice were used to construct acute cerebral infarction model. The model mice were randomly divided into model group and experimental group according to their body weight. The experimental group received 20 mg kg~ (-1) ginkgo biloba flavone intraperitoneally every day. The sham operation group and the model group were given 0.9 NaCl3 for 7 days. The expression levels of HIF-1 偽, TNF- 偽 and Caspase-3 in serum of the mice with acute cerebral infarction were detected by enzyme-linked immunosorbent assay (Elisa). Results the levels of TNF- 偽 in serum of sham operation group, model group and experimental group were (5.16 鹵1.04), (33.02 鹵20.34), (22.64 鹵10.17) ng mL~ (-1); The HIF-1 偽 in serum of the three groups was (508.94 鹵123.18), () 735.19 鹵324.05), (626.24 鹵212.76) ng mL~ (-1); The Caspase-3 in serum of the three groups was (6.54 鹵0.24), (12.23 鹵3.45), (10.24 鹵1.58) ng mL~ (-1); The expression of TNF- 偽, HIF-1 偽 and Caspase-3 in serum of model group was significantly higher than that of sham operation group (P0.05). The expression of TNF- 偽, HIF-1 偽 and Caspase-3 in serum of experimental group was significantly lower than that of model group (P0.05). Conclusion Ginkgo biloba flavonoids can significantly decrease the levels of TNF- 偽, Caspase-3 and HIF-1 偽 in serum of mice with acute cerebral infarction.
