【摘要】：目的:克隆表達(dá)家蠶(Bombyx mori)Profilin蛋白,鑒定其免疫原性并進(jìn)行B細(xì)胞抗原表位預(yù)測(cè)和構(gòu)建分子進(jìn)化樹。方法:從NCBI上獲取家蠶Profilin蛋白的基因序列,合成該基因并構(gòu)建到pet-28a表達(dá)載體上,重組質(zhì)粒轉(zhuǎn)化至E.coli BL21,IPTG誘導(dǎo)基因表達(dá),通過親和層析獲取高純度蛋白;用Western blot方法對(duì)重組蛋白進(jìn)行過敏原性鑒定;通過DNAStar軟件分析其潛在的B細(xì)胞抗原表位;應(yīng)用MEGA5.05進(jìn)行序列比對(duì),并構(gòu)建分子進(jìn)化樹。結(jié)果:成功表達(dá)出重組蛋白,且重組蛋白與家蠶過敏患者血清有一定的Ig E結(jié)合。結(jié)論:成功克隆表達(dá)出具有免疫原性的Profilin蛋白,并成功預(yù)測(cè)出其B細(xì)胞抗原表位和構(gòu)建出分子進(jìn)化樹。
[Abstract]:Objective: to clone and express (Bombyx mori) Profilin protein of Bombyx mori, identify its immunogenicity, predict the epitope of B cell antigen and construct a molecular evolutionary tree. Methods: the gene sequence of Profilin protein of Bombyx mori (Bombyx mori) was obtained from NCBI. The gene was synthesized and constructed into pet-28a expression vector. The recombinant plasmid was transformed into E.coli BL21,IPTG induced gene expression, and high purity protein was obtained by affinity chromatography. Western blot method was used to identify the allergen of recombinant protein, DNAStar software was used to analyze the potential B cell epitopes, MEGA5.05 was used to sequence alignment, and molecular evolutionary tree was constructed. Results: the recombinant protein was successfully expressed, and the recombinant protein had certain Ig E binding to the sera of silkworm allergy patients. Conclusion: Profilin protein with immunogenicity was cloned and expressed successfully, and its B cell epitopes were successfully predicted and molecular evolutionary tree was constructed.
