補陽還五湯聯合MSCs移植對TIMP-1干預后MCAO模型大鼠凝血Ⅷ因子的影響

【摘要】:目的:揭示補陽還五湯聯合MSCs移植對大腦中動脈栓塞再灌注大鼠模型((MCAO))抗凝血、促進血管新生的機理。方法:采用TIMP-1對模型進行干預,側腦室給予MSCs,聯合補陽還五湯灌胃,采用免疫組化法檢測凝血因子Ⅷ的表達。結果:聯合補陽還五湯聯合MSCs移植組能夠降低缺血區(qū)凝血因子Ⅷ數量,12 h、24 h最為明顯,與模型組、抑制劑組、單純MSCs組相比,有極顯著性差異(P0.01);至36 h時有一定升高,與其他組相比,無顯著性差異(P0.05),隨后表達Ⅷ因子數量又開始下降,至48 h時,與模型組、抑制劑組相比,有顯著性差異(P0.05),與單純MSCs組相比,無顯著性差異(P0.05)。結論:補陽還五湯聯合MSC移植,能夠降低TIMP-1干預后腦缺血模型大鼠缺血腦組織Ⅷ因子數量,起到抗凝血,抑制血栓的形成。
