黃芩苷對(duì)潰瘍性結(jié)腸炎模型大鼠NF-κB表達(dá)的影響

【摘要】:目的觀察黃芩苷對(duì)潰瘍性結(jié)腸炎模型大鼠核因子?κB(Nuclear factor?κB,NF-κB)表達(dá)及下游炎癥細(xì)胞因子的影響。方法采用TNBS法制備大鼠潰瘍性結(jié)腸炎模型,隨機(jī)分為正常組、模型組、黃芩苷(Baicalin)高中低劑量組、陽(yáng)性對(duì)照組(柳氮磺胺吡啶,SASP組)。給藥10d后,留取結(jié)腸組織標(biāo)本,觀察結(jié)腸大體形態(tài)變化,采用ELISA法觀察UC模型大鼠結(jié)腸組織中腫瘤壞死因子(TNF-α)、白介素-8(IL-8)和白介素-1(IL-1)的含量,采用Western blot法檢測(cè)NF-κB的表達(dá)水平。結(jié)果與模型組相比,黃芩苷100mg/kg組能顯著改善結(jié)腸大體形態(tài),黃芩苷(25、50、100mg/kg)干預(yù)后能劑量依賴性的抑制各炎癥因子的分泌,黃芩苷高劑量組能降低IL-1、IL-8和TNF-α表達(dá)水平,但與SASP組比較無(wú)明顯差異。黃芩苷高劑量組能降低NF-κB的水平,促進(jìn)IκBα水平(P0.05)。結(jié)論黃芩苷能夠抑制NF-κB的活化,從而發(fā)揮其在潰瘍性結(jié)腸炎中的抗炎效應(yīng)。
[Abstract]:Objective to observe the effect of baicalin on the expression of nuclear factor-魏 B (Nuclear factor? 魏 B-魏 NF- 魏 B and inflammatory cytokines in rats with ulcerative colitis. Methods the ulcerative colitis model of rats was established by TNBS method. The rats were randomly divided into normal group, model group, baicalin (Baicalin) high and low dose group, and positive control group (SASP group). After 10 days of administration, the colonic tissue samples were collected to observe the changes of colonic gross morphology, and the tumor necrosis factor (TNF- 偽) in colonic tissue of UC model rats was observed by ELISA method. The levels of interleukin-8 (IL-8) and interleukin-1 (IL-1) were detected by Western blot assay. Results compared with the model group, baicalin 100mg/kg group could significantly improve the colonic gross morphology. Baicalin (2550 mg / kg) could inhibit the secretion of inflammatory factors in a dose-dependent manner, while baicalin high dose group could reduce IL-1,. The expression levels of IL-8 and TNF- 偽 were not significantly different from those of SASP group. Baicalin high dose group could decrease the level of NF- 魏 B and promote the level of I 魏 B 偽 (P0.05). Conclusion baicalin can inhibit the activation of NF- 魏 B and exert its anti-inflammatory effect in ulcerative colitis.
