電針刺激對脊髓挫傷模型大鼠神經(jīng)細胞凋亡的影響

【摘要】:背景:實驗研究表明,電針對于脊髓損傷的功能恢復(fù)具有促進作用,可以降低疼痛,提高急性脊髓損傷的術(shù)后療效。目的:觀察脊髓挫傷造模后電針刺激干預(yù)對損傷區(qū)細胞凋亡的影響,并分析其對脊髓挫傷大鼠神經(jīng)功能保護的可能作用機制。方法:成年雌性SD大鼠66只,采用隨機區(qū)組法分為假手術(shù)組20只和大鼠急性脊髓挫傷造模成功40只,造模成功大鼠再隨機分為脊髓挫傷組、電針刺激組。分別于造模前、造模后1 d、3 d、1周、2周、3周、4周通過BBB評分、斜板試驗進行運動功能評定。造模后3 d,通過TUNEL法檢測各實驗組大鼠脊髓挫傷區(qū)神經(jīng)細胞的凋亡情況;RT-PCR、Western blot檢測脊髓挫傷區(qū)周圍bax、bcl-2、caspase-3基因轉(zhuǎn)錄和蛋白的表達;蘇木精-伊紅染色觀察脊髓挫傷區(qū)病理組織形態(tài)學(xué)變化;HRP示蹤觀察神經(jīng)纖維的再生情況。結(jié)果與結(jié)論:1電針刺激組2項運動功能評分在治療后2周各個時間點均較脊髓挫傷組顯著增加(P0.05)。2造模后3 d經(jīng)脊髓電針刺激組細胞凋亡指數(shù)均明顯低于脊髓挫傷組(P0.05)。3造模后72 h,與脊髓挫傷組相比經(jīng)脊髓電針刺激組bax、caspase-3基因和蛋白表達顯著降低(P0.05);bcl-2基因和蛋白表達顯著升高(P0.05)。4造模后4周,HRP陽性神經(jīng)纖維數(shù):假手術(shù)組最多,電針刺激組次之,脊髓挫傷組最少,各組之間差異有顯著性意義(P0.05)。結(jié)果提示,電針刺激治療通過減少大鼠脊髓挫傷區(qū)神經(jīng)細胞的凋亡對脊髓挫傷起到保護作用。
[Abstract]:Background: experimental studies have shown that electroacupuncture can promote the functional recovery of spinal cord injury, can reduce pain and improve the postoperative efficacy of acute spinal cord injury. Aim: to observe the effect of electroacupuncture (EA) stimulation on cell apoptosis after spinal cord contusion in rats, and to analyze the possible mechanism of its protective effect on spinal cord contusion in rats. Methods: 66 adult female SD rats were randomly divided into sham operation group (n = 20) and acute spinal cord contusion model (n = 40). The motor function was evaluated by BBB score and inclined plate test before and 1 day after model making and 1 week, 2 weeks, 3 weeks and 4 weeks after modeling. The apoptosis of neurons in the contusion area of spinal cord was detected by TUNEL method and the expression of bax,bcl-2,caspase-3 gene and protein around the contusion area of spinal cord was detected by RT-PCR,Western blot on the 3rd day after the establishment of the model. Histopathological changes in contusion of spinal cord were observed by hematoxylin-eosin staining and regeneration of nerve fibers was observed by HRP tracer. Results and conclusion: 1 the motor function scores of the electroacupuncture group were significantly higher than those of the spinal cord contusion group at every time point 2 weeks after treatment (P0.05). 2 the apoptotic index of the electroacupuncture stimulation group was significantly higher than that of the spinal cord contusion group 3 days after the treatment (P0.05). It was significantly lower than that in the spinal cord contusion group (P0.05). Compared with the spinal cord contusion group, the expression of bax,caspase-3 gene and protein decreased significantly in the spinal cord electroacupuncture group (P0.05). The expression of bcl-2 gene and protein increased significantly (P0.05). 4 the number of HRP positive nerve fibers was the most in sham operation group, followed by electroacupuncture stimulation group and spinal cord contusion group (P0.05). 4. 4 weeks after modeling, the number of HRP positive nerve fibers was the highest in sham operation group, second in electroacupuncture group, and the least in spinal cord contusion group (P0.05). The results suggest that electroacupuncture therapy can protect spinal cord contusion by reducing the apoptosis of nerve cells in spinal cord contusion area.
