不同濃度彈力蛋白酶對(duì)建立兔腹主動(dòng)脈瘤模型成瘤率的影響

【摘要】:目的通過腹主動(dòng)脈腔內(nèi)低壓力灌注豬胰彈力蛋白酶建立兔腹主動(dòng)脈瘤模型,探索不同濃度豬胰彈力蛋白酶對(duì)兔腹主動(dòng)脈成瘤率的影響。方法新西蘭雄兔50只,體質(zhì)量(2.50±0.20)kg,隨機(jī)分為5組:A組、B組、C組、D組、E組,每組各10只。A組經(jīng)雙腔導(dǎo)管注入生理鹽水10μl,其余4組經(jīng)雙腔導(dǎo)管注入10μl濃度分別為1U/ml、10U/ml、100U/ml、200U/ml的豬胰彈力蛋白酶,灌注保持7min。術(shù)后第7天處死動(dòng)物,分別于術(shù)前及術(shù)后第7天行腹主動(dòng)脈彩超檢查,觀察灌注血管部位變化。結(jié)果 E組實(shí)驗(yàn)動(dòng)物存活率較A組、B組、C組及D組明顯下降(40%vs 90%、90%、90%及90%)。腹主動(dòng)脈術(shù)后最大直徑隨給予灌注濃度增加而增大。與術(shù)前最大直徑比較,A組及B組術(shù)后最大直徑未見明顯擴(kuò)張;C組、D組及E組術(shù)后最大直徑明顯擴(kuò)張;D組及E組較A組、B組及C組,術(shù)后第7天腹主動(dòng)脈擴(kuò)張率[(125.40±16.10)%及(248.20±5.60)%vs 0%、(24.90±2.50)%及(43.90±7.69)%,P0.05]及成瘤率[100.0%及100.0%vs 0%、0%及33.3%,P0.05]均明顯升高。結(jié)論高濃度豬胰彈力蛋白酶(100U/ml)在低壓力狀態(tài)下較短時(shí)間灌注可成功建立兔腹主動(dòng)脈瘤模型,且成瘤率高、死亡率低。
[Abstract]:Objective to establish a rabbit model of abdominal aortic aneurysm by perfusion of porcine pancreatic elastase into abdominal aorta under low pressure, and to explore the effect of different concentrations of porcine pancreatic elastase on the rate of abdominal aortic aneurysm formation in rabbits. Methods Fifty New Zealand male rabbits with a body weight of (2.50 鹵0.20) kg, were randomly divided into five groups: group A, group B, group C, group D and group E. The other four groups were infused with porcine pancreatic elastase at the concentration of 1 U / ml 10 U / ml 100 U / ml / ml through double lumen catheter for 7 min. The animals were killed on the 7th day after operation. The abdominal aorta was examined by color Doppler ultrasonography before operation and on the 7th day after operation. Results the survival rate of experimental animals in group E was significantly lower than that in group A, group B, group C and group D (40%vs 90% and 90%). The maximum diameter of abdominal aorta increased with the increase of perfusion concentration. The maximum diameter of group A and group B was not significantly expanded compared with that before operation, and that of group C, group D and group E was significantly expanded after operation. The abdominal aortic dilatation rates in group D and E were (125.40 鹵16.10)% and (248.20 鹵5.60)% vs _ 0, (24.90 鹵2.50)% and (43.90 鹵7.69)% respectively on the 7th day after operation, compared with those in group A, B and C [(125.40 鹵16.10)% and (248.20 鹵5.60)%, respectively]. The rate of tumorigenesis [100.0%, 100.0%vs 0% and 33.3P] were significantly higher than that of control group (P 0.05). Conclusion High concentration porcine pancreatic elastase (100U/ml) can successfully establish rabbit abdominal aortic aneurysm model under low pressure for a short time, and the tumorigenesis rate is high and the mortality is low.
