【摘要】：為了運用噬菌體展示技術(shù)對抗白細(xì)胞介素-4受體(IL-4R)特異性單鏈抗體進(jìn)行表達(dá)與鑒定,試驗提取了前期工作中經(jīng)富集與篩選抗IL-4R單鏈抗體庫而得到的特異性pCANTAB5E-sc Fv重組質(zhì)粒,用NotⅠ和SfiⅠ雙酶切鑒定樣本并將質(zhì)粒送上海英駿生物公司測序;利用pCANTAB-5E噬菌體展示系統(tǒng)(pCANTAB-5E、TG1、M13K07、HB2151)表達(dá)抗IL-4R單鏈抗體重組蛋白;通過SDS-PAGE和Western-blot分析單鏈抗體的表達(dá)情況及抗體的活性。結(jié)果表明:pCANTAB-5E噬菌體展示系統(tǒng)表達(dá)出的單鏈抗體重組蛋白分子質(zhì)量在25~35 ku之間,28 ku左右,與測序結(jié)果預(yù)測的蛋白分子質(zhì)量接近。說明成功表達(dá)了抗IL-4R單鏈抗體重組蛋白。
