【摘要】：為研究棕櫚�；瘜χ行粤＜毎吇缘恼{(diào)節(jié)作用,利用2-溴棕櫚酸(2-bromopalmitate,2-BP)或棕櫚酸(palmitate)預處理來源于骨髓的中性粒細胞.采用流式細胞術(flow cytometry,FCM)檢測中性粒細胞的存活率和中性粒細胞內(nèi)F-肌動蛋白(F-actin)的生成;通過免疫熒光分析中性粒細胞內(nèi)F-actin的極化;利用細胞遷移試驗檢測中性粒細胞的遷移能力;通過免疫印跡的實驗方法檢測蛋白激酶B(protein kinase B,AKT)和磷酸化AKT的表達.結果發(fā)現(xiàn),2-BP預處理顯著抑制了中性粒細胞內(nèi)F-actin極化,且抑制了中性粒細胞向趨化肽N-甲酰甲硫氨酰-亮氨酰-苯丙氨酸(N-formylmethionyl-leucyl-phenylalanine,f MLP)遷移的能力,而棕櫚酸預處理顯著促進了中性粒細胞內(nèi)F-肌動蛋白極化.兩者對F-actin生成和AKT的磷酸化均無影響.研究結果表明,2-BP處理可能通過影響F-actin的極化抑制了中性粒細胞向f MLP的趨化活性.
[Abstract]:In order to study the regulation of palmitoacylation on neutrophil chemotaxis, 2-bromopalmitate 2-BP (2-bromopalmitate 2-BP) or palmitic acid (palmitate) were pretreated from neutrophils from bone marrow. Flow cytometry (flow cytometry,FCM) was used to detect the survival rate of neutrophils and the production of F-actin (F-actin) in neutrophils, and the polarization of F-actin in neutrophils was analyzed by immunofluorescence. The translocation ability of neutrophils was detected by cell migration assay, and the expression of protein kinase (B (protein kinase) and phosphorylated AKT was detected by Western blotting. The results showed that 2-BP pretreatment significantly inhibited the F-actin polarization in neutrophils and the ability of neutrophils to migrate to N-formylmethionyl-leucyl-phenylalanine (MLP). Palmitic acid pretreatment significantly promoted F-actin polarization in neutrophils. Both of them had no effect on F-actin production and AKT phosphorylation. The results show that 2-BP treatment may inhibit the chemotaxis of neutrophils to f MLP by affecting the polarization of F-actin.
