旋毛蟲病人血清Western blot對成蟲可溶性蛋白中早期診斷抗原的鑒定

【摘要】:目的鑒定旋毛蟲成蟲早期特異性診斷抗原。方法應(yīng)用感染19d和35d的旋毛蟲病患者血清,通過Western blot對旋毛蟲成蟲可溶性蛋白的抗原組分進行篩選和鑒定。結(jié)果 SDS-PAGE顯示,旋毛蟲成蟲可溶性蛋白有27條蛋白帶,分子質(zhì)量單位分別為95、81.3、73.4、63.6、61.4、55、50.6、47、45.1、43.7、41.9、40.6、39、37.4、35.1、33.3、31、30.4、27.8、26.6、25.4、24、21、19.2、17.9、15.4和14.4ku。Western blot顯示,成蟲可溶性蛋白中的41.9ku和50.6ku蛋白與感染19d和35d的旋毛蟲病患者血清反應(yīng)率均為100%(15/15),與囊尾蚴病、裂頭蚴病、并殖吸蟲病、華支睪吸蟲病患者血清及健康人血清均無交叉反應(yīng),與棘球蚴病和日本血吸蟲病患者血清的交叉反應(yīng)率分別為23.8%和44.44%。結(jié)論旋毛蟲成蟲可溶性蛋白中的41.9ku和50.6ku蛋白用于人體旋毛蟲病早期診斷具有高度的敏感性和良好的特異性。
[Abstract]:Objective to identify the early specific diagnostic antigen of Trichinella spiralis adult. Methods the antigen components of soluble protein of adult Trichinella spiralis were screened and identified by Western blot in sera of patients infected with trichinellosis for 19 and 35 days. Results SDS-PAGE showed that there were 27 protein bands in soluble protein of adult Trichinella spiralis. The molecular weight units are 95U 81.3C 73.4N 63.6N 61.4U 50.6U 45.6N 45.7U 41.9U 40.6N 41.9U 40.6N 39.39.36.31.31.31.31.31.31.31.31.31.33.3cu 33.826.625.42421,19.2hu 17.9 / 15.4, respectively, according to the results of 14.4ku.Western blot, the molecular weight units of the two groups are as follows: (1) the molecular weight units are:! 27. 826! 36.625. The response rates of 41.9ku and 50.6ku proteins in soluble proteins of adult worms were 100% (15 / 15) in sera of patients infected with trichinellosis for 19 and 35 days, and were associated with cysticercosis, cysticercosis and paragonimiasis. There was no cross reaction in sera of clonorchiasis patients and healthy persons. The cross reaction rates with sera of patients with echinococcosis and schistosomiasis japonicum were 23.8% and 44.44%, respectively. Conclusion 41.9ku and 50.6ku proteins in soluble protein of adult Trichinella spiralis have high sensitivity and good specificity for early diagnosis of human trichinellosis.
