【摘要】：目的:探討補腎活血化痰湯對多囊卵巢綜合征(PCOS)模型大鼠卵巢血供及形態(tài)的影響。方法:取SD大鼠(♀)50只隨機分為正常對照組、模型組、陽性對照組[羅格列酮,3 mg/(kg·d)]和補腎活血化痰湯低、高劑量組[5.0、7.5 g/(kg·d)],每組10只。除正常對照組外,其余各組大鼠均采用Poresky法復(fù)制PCOS模型。成模后,各給藥組大鼠ig相應(yīng)藥物,正常對照組和模型組大鼠ig生理鹽水[10 m L/(kg·d)],每天1次,連續(xù)22 d。給藥結(jié)束后處死大鼠,記錄微血管密度(MVD),檢測大鼠卵巢組織血管內(nèi)皮生長因子(VEGF)、血管緊張素Ⅱ(AngⅡ)1型受體(AT1)和2型受體(AT2)以及G蛋白偶聯(lián)受體MAS m RNA的表達,并觀察卵巢形態(tài)變化。結(jié)果:與正常對照組比較,模型組和各給藥組大鼠MVD均升高,AT1、AT2、MAS m RNA表達均下調(diào)(P0.05);模型組大鼠卵巢組織可見囊性竇卵泡、顆粒細(xì)胞層減少,卵母細(xì)胞消失,黃體組織數(shù)量減少等明顯病理變化。與模型組比較,各給藥組上述指標(biāo)均得到顯著改善,且補腎活血化痰湯低、高劑量組作用均強于陽性對照組(P0.05);各給藥組大鼠的卵巢組織病理變化均得到一定改善。結(jié)論:補腎活血化痰湯可增加PCOS模型大鼠卵巢組織血供,促進血管生成,促排卵,有助于改善卵巢形態(tài)。
