人與獼猴不同組織多能性干細(xì)胞相關(guān)基因和標(biāo)志抗原的表達(dá)分析

【摘要】:近來(lái)研究表明臍帶、臍帶血、羊水以及出生后外周血液中存在具有多分化潛能的干細(xì)胞,但研究結(jié)果不盡一致;Oct-4、Nanog、Sox-2、Rex-1、SSEAs作為胚胎干細(xì)胞的標(biāo)志分子,常用于評(píng)價(jià)干細(xì)胞的多潛能性。本研究采用半定量RT-PCR法分析多能性干細(xì)胞相關(guān)基因Oct-4、Nanog、Sox-2以及Rex-1在人源臍帶、臍帶血、羊水組織以及獼猴外周血液中的表達(dá)情況,并對(duì)上述基因的擴(kuò)增片段進(jìn)行同源分析;同時(shí)用SP免疫組織化學(xué)法分析Oct-4、Nanog、Sox-2、SSEA-1、SSEA-3、SSEA-4在人源臍帶組織中的表達(dá)和分布情況,其結(jié)果如下: 1、Oct-4、Nanog、Sox-2以及Rex-1基因在人源臍帶、臍帶血、羊水組織中均穩(wěn)定表達(dá)(陽(yáng)性率≥80%);在上述組織中,Oct-4、Nanog, Sox-2以及Rex-1基因的表達(dá)水平存在差異,其在臍帶和羊水中表達(dá)水平差異不顯著(P0.05),但二者都顯著高于臍帶血(P0.05) 2、在不同生長(zhǎng)階段健康獼猴外周血液中Oct-4、Nanog和Sox-2基因均呈陽(yáng)性表達(dá)(陽(yáng)性表達(dá)率≥80%);Oct-4、Nanog和Sox-2基因在獼猴外周血液中的表達(dá)水平與年齡呈負(fù)相關(guān),但差異不顯著(P0.05)。 3、將人源臍帶、臍帶血、羊水、獼猴外周血組織中擴(kuò)增所得Oct-4、Nanog、Sox-2基因序列,與人、獼猴、豬、牛、小鼠、褐家鼠、狗、兔等部分物種上述基因cDNA序列同源性比較,結(jié)果顯示,人與獼猴、兔、豬、牛有較高同源性(75%-95.9%),與小鼠和褐家鼠的同源性60%以上;上述基因序列生物進(jìn)化分析的結(jié)果顯示,克隆所得目的基因序列與人、獼猴相同基因的序列親緣關(guān)系最近,三者構(gòu)成親緣關(guān)系最近的一個(gè)分支;再與豬、牛和狗的該基因構(gòu)成一個(gè)較大的分支,其次親緣關(guān)系依次為兔、大鼠和小鼠等。 4、在人源臍帶組織中,均檢測(cè)到表達(dá)Oct-4、Nanog、Sox-2、SSEA-1、SSEA-3、SSEA-4等標(biāo)記分子的陽(yáng)性細(xì)胞,陽(yáng)性(陽(yáng)性程度大于或等于(++))表達(dá)率在75%以上,且各標(biāo)記分子陽(yáng)性表達(dá)率差異不顯著(P0.05),這些細(xì)胞主要分布于臍帶組織的華爾通膠和臍血管周圍,但血管內(nèi)皮附近分布較少。
[Abstract]:Recent studies have shown that there are stem cells with multiple differentiation potential in umbilical cord, umbilical cord blood, amniotic fluid and postnatal peripheral blood, but the results are not consistent. As a marker of embryonic stem cells, Oct-4,Nanog,Sox-2,Rex-1,SSEAs is often used to evaluate the pluripotency of stem cells. The expression of Oct-4,Nanog,Sox-2 and Rex-1 in human umbilical cord, umbilical cord blood, amniotic fluid and peripheral blood of rhesus monkey were analyzed by semi-quantitative RT-PCR. The amplified fragments of the above genes were analyzed by homology analysis. At the same time, the expression and distribution of Oct-4,Nanog,Sox-2,SSEA-1,SSEA-3,SSEA-4 in human umbilical cord tissues were analyzed by SP immunohistochemical method.
