【摘要】：目的 研究J774細(xì)胞凋亡與PA上清液感染的關(guān)系；應(yīng)用western blot法研究Fas和FAP-1蛋白在PA上清液誘導(dǎo)J774細(xì)胞凋亡過程中的作用。 方法 1、用15%濃度的PA上清液感染J774細(xì)胞3h，6h，9h，，在37℃培養(yǎng)箱中培養(yǎng)。 2、以Annexin-V-FITC/PI雙染流式細(xì)胞儀檢測及Hoechest33258熒光染色等細(xì)胞形態(tài)學(xué)觀察指標(biāo)，研究PA上清液感染對J774細(xì)胞凋亡的誘導(dǎo)作用。 3、用western blot法對FAP-1和Fas蛋白定量檢測。 結(jié)果 Hoechest33258熒光顯微鏡下觀察：15%濃度的PA上清液感染J774細(xì)胞9h可見許多細(xì)胞有典型的凋亡形態(tài)學(xué)改變，并可見凋亡小體。經(jīng)Hoechest33258熒光染色檢查和Annexin-V-FITC/PI雙染流式細(xì)胞儀檢測，隨著PA上清液感染J774細(xì)胞時間的延長，J774細(xì)胞凋亡率均增高。各實(shí)驗(yàn)組J774細(xì)胞凋亡率均數(shù)與對照組比較差異均有統(tǒng)計學(xué)意義（P0.01）；各實(shí)驗(yàn)組間比較差異均有統(tǒng)計學(xué)意義（P0.01）；兩種檢測方法比較，相同時間感染條件下的J774細(xì)胞凋亡率均數(shù)差異無統(tǒng)計學(xué)意義（P0.05）。western blot結(jié)果顯示，在PA上清液感染J774細(xì)胞3h后，F(xiàn)as蛋白表達(dá)與對照組相比明顯增強(qiáng)，且隨著時間的延長表達(dá)逐漸增強(qiáng)，直到PA上清液感染J774細(xì)胞9h；而FAP-1蛋白表達(dá)隨著時間的延長表達(dá)逐漸減弱。 結(jié)論 1、J774巨噬細(xì)胞凋亡與銅綠假單胞菌上清液感染有關(guān)。 2、銅綠假單胞菌上清液導(dǎo)致的J774細(xì)胞凋亡具有時間依賴性。 3、該實(shí)驗(yàn)凋亡的機(jī)制涉及Fas和FAP-1蛋白的表達(dá)。
