HIF-1α在失血性動物心肌中的表達
[Abstract]:Aim: to investigate the expression and significance of hypoxia inducible factor-1 (hypoxia-inducible factor-1) protein and mRNA in cardiac myocytes of rats with different degrees of blood loss. Methods: blood loss models were established in clean grade SD rats. The rats were randomly divided into three groups: (CON), 10% (10-BL), bleeding 20% (20-BL), and bleeding 30% (30-BL). The cardiac function was monitored by Powerlab eight-channel physiological recorder, and the expression of HIF-1 m RNA and protein was detected by RT-PCR and Western Blot in the myocardium of rats 30 min after blood loss. The distribution of HIF-1 in cardiomyocytes was detected by immunohistochemical staining. Results: compared with CON group, the expression of HIF-1 mRNA in 10-BL20-BL 30-BL group was increased gradually (P0.05), and the results of RT-PCR semi-quantitative analysis showed that the cardiac function decreased significantly at 1 hour after different degree of blood loss (P 0.05), and the expression of HIF-1 mRNA in 10-BL20-BL 30-BL group was significantly higher than that in CON group (P0.05). Immunohistochemical results showed that HIF-1 偽 was mainly distributed in the cytoplasm of myocardium and also in the nucleus. Conclusion: with the increase of blood loss, the expression of HIF-1 偽 is increased. HIF-1 偽 may be involved in the adaptive regulation of myocardial metabolism in hypoxic state induced by blood loss. Methods: a blood loss model was established in small white pigs. The model was randomly divided into three groups: 10% (10-BL), 20% (20-BL), and 30% (30-BL) of (CON), in control group. The cardiac function was monitored by Powerlab eight-channel physiological recorder. The expression of HIF-1 mRNA and protein was detected by RT-PCR and Western Blot, and the distribution of HIF-1 in cardiac myocytes was detected by immunohistochemical staining. Results: after different degree of blood loss, cardiac function index decreased, RT-PCR semi-quantitative results showed that compared with CON group, the expression of HIF-1 mRNA increased gradually in 10-BL20-BL 30-BL group (P0.05). Immunohistochemical results showed that HIF-1 偽 was mainly distributed in the cytoplasm of myocardium and also in the nucleus. Conclusion: the degree of blood loss is increased, the time is prolonged and the expression of HIF-1 偽 is increased. HIF-1 偽 may be involved in the adaptive regulation of myocardial metabolism in hypoxic state induced by blood loss.
