高效檢測人降鈣素原化學(xué)發(fā)光免疫分析方法的建立與評價

【摘要】:目的重組原核表達(dá)人降鈣素原(PCT)蛋白,制備抗人PCT單克隆抗體,建立高效檢測PCT化學(xué)發(fā)光免疫分析方法。方法根據(jù)NCBI提供的PCT基因序列,按照大腸埃希菌偏愛密碼子優(yōu)化后進(jìn)行全基因合成,通過BamHⅠ和XhoⅠ雙酶切將其構(gòu)建到pET32a載體上;將pET32a-PCT質(zhì)粒轉(zhuǎn)化至大腸埃希菌BL21(DE3),用異丙基-β-D-硫代半乳糖苷(IPTG)誘導(dǎo)表達(dá),采用Western blot法分析重組蛋白表達(dá)情況;用鎳親和純化柱純化重組蛋白,以此為抗原免疫BALB/c小鼠。取免疫小鼠脾細(xì)胞與小鼠骨髓瘤SP2/0細(xì)胞融合,采用間接ELISA法篩選陽性雜交瘤細(xì)胞,有限稀釋法進(jìn)行克隆化后再用間接ELISA法篩選陽性單克隆細(xì)胞;將陽性單克隆細(xì)胞接種于小鼠腹腔內(nèi),制備腹水單抗,經(jīng)Protein A/G純化后進(jìn)行十二烷基硫酸鈉-聚丙烯酰胺凝膠電泳(SDS-PAGE)鑒定,采用間接ELISA方法對抗體的特異性、靈敏度以及親和力進(jìn)行鑒定;用NaIO4氧化HRP法標(biāo)記單克隆抗體,通過棋盤法篩選配對單克隆抗體,以篩選的配對抗體為捕捉抗體,建立雙抗夾心化學(xué)發(fā)光免疫分析的血清學(xué)檢測體系,檢測臨床150例血清標(biāo)本,其中PCT升高(0.05ng/ml)120例,PCT陰性(0.05ng/ml)30例。結(jié)果共篩選出6株抗人PCT蛋白的單克隆細(xì)胞,制備的腹水效價均大于4×10~(-6);通過棋盤法篩選得到2對能夠進(jìn)行雙抗夾心配對的單抗,其中1對親和力較高,其親和力常數(shù)分別為2.39×10~8 L/mol和2.91×10~8 L/mol。雙抗夾心化學(xué)發(fā)光免疫分析方法檢測線性范圍為0.06~8ng/ml,其中陽性檢出率為96.6%,陰性符合率為100%,與臨床檢測數(shù)據(jù)相比,相關(guān)系數(shù)R~2=0.9737。結(jié)論建立的雙抗夾心ELISA方法靈敏度高、特異性強、穩(wěn)定可靠,可用于細(xì)菌、寄生蟲等病原體感染患者的PCT血清學(xué)定量檢測。
