【摘要】：乙肝核心抗原(HBcAg)具有天然形成病毒樣顆粒的能力,同時這種結(jié)構(gòu)能使機體產(chǎn)生特異性的體液免疫和細胞免疫作用,故成為了良好的免疫載體蛋白。本實驗用含有231個氨基酸的大分子礁珊瑚熒光蛋白(ZsGreen)作為的目標抗原,以具備顆粒裝配區(qū)的乙肝核心抗原蛋白為抗原遞呈的載體,通過基因克隆技術(shù)將表達ZsGreen蛋白的基因插入HBcAg中位于顆粒表面的主要免疫優(yōu)勢抗原決定簇c/e1表位區(qū),并將這部分融合基因與載體pET28a進行連接,構(gòu)建重組表達載體。重組表達載體轉(zhuǎn)化至大腸桿菌BL21(DE3),經(jīng)過對表達條件及發(fā)酵工藝條件的優(yōu)化后,重組融合蛋白(Zscore)在大腸桿菌中的表達量達到20%以上。表達菌體經(jīng)過破璧,離心澄清,硫酸銨沉淀,陽離子及分子篩層析等方法的分離純化,最終獲得顆粒純度大于80%的重組融合蛋白。重組融合蛋白經(jīng)過系統(tǒng)的結(jié)構(gòu)分析及免疫學研究,證實:(1)HBcAg在融合大分子蛋白ZsGreen后其產(chǎn)物仍能形成病毒樣顆粒(VLP)的結(jié)構(gòu),(2)且展示在重組蛋白顆粒表面融合表達的ZsGreen能激發(fā)更高的免疫原性。這一系列的證據(jù)表明乙肝核心抗原展示表達外源蛋白的平臺具備疫苗表達載體的能力,為進一步的研究開發(fā)打下基礎(chǔ)。
[Abstract]:Hepatitis B core antigen (HBcAg) has the ability of forming virus-like particles naturally, and this structure can make the body produce specific humoral and cellular immunity, so it has become a good immune carrier protein. In this experiment, the fluorescent protein (ZsGreen) of reef coral containing 231 amino acids was used as the target antigen, and the hepatitis B core antigen protein with granular assembly region was used as the antigen presenting carrier. The gene expressing ZsGreen protein was inserted into the c/e1 epitope of the main immune dominant antigen determinant on the surface of particles in HBcAg by gene cloning technique. The fusion gene was ligated with the vector pET28a to construct the recombinant expression vector. The recombinant expression vector was transformed into Escherichia coli BL21 (DE3). After optimizing the expression conditions and fermentation conditions, the recombinant fusion protein (Zscore) was expressed more than 20% in Escherichia coli. The recombinant fusion protein with particle purity of more than 80% was obtained by the separation and purification of the expressed bacteria through the separation and purification of the expressed bacteria by centrifugation, ammonium sulfate precipitation, cationic and molecular sieve chromatography and so on. The recombinant fusion protein was studied by systematic structural analysis and immunological analysis. The results showed that: (1) HBcAg could still form the structure of virus-like (VLP) after fusion of macromolecular protein ZsGreen, and (2) ZsGreen expressed on the surface of recombinant protein particles could stimulate higher immunogenicity. This series of evidences indicate that hepatitis B core antigen display the ability to express foreign protein on the platform of vaccine expression vector and lay the foundation for further research and development.
