干酪乳酸桿菌代謝物對(duì)白假絲酵母菌生物被膜的體外抑菌效果及分子機(jī)制

【摘要】:目的研究干酪乳酸桿菌代謝物對(duì)體外白假絲酵母菌生物被膜的抑菌作用及其機(jī)制。方法通過萃取、蒸餾的方式濃縮干酪乳酸桿菌代謝物。采用甲基四氮鹽(XTT)減低法測(cè)定干酪乳酸桿菌代謝物對(duì)白假絲酵母菌YEM30游離態(tài)及生物被膜態(tài)的最小抑菌濃度(MIC80),定量檢測(cè)干酪乳酸桿菌代謝物對(duì)白假絲酵母菌體外代謝活動(dòng)的影響,觀察白假絲酵母菌形態(tài)學(xué)的轉(zhuǎn)變,并通過雙向凝膠電泳(2-DE)對(duì)比干酪乳酸桿菌代謝物處理前后白假絲酵母菌蛋白的表達(dá)差異。結(jié)果干酪乳酸桿菌代謝物對(duì)白假絲酵母菌的游離態(tài)、生物被膜態(tài)的最小抑菌稀釋倍數(shù)分別為1∶256和1∶128。稀釋倍數(shù)為1∶256的干酪乳酸桿菌代謝物可以明顯干擾白假絲酵母菌酵母相向菌絲相的轉(zhuǎn)化。比較雙向電泳圖譜上獲得6個(gè)顯著蛋白斑點(diǎn),對(duì)其中的4個(gè)進(jìn)行鑒定,結(jié)果顯示在干酪乳酸桿菌代謝物的刺激下,烯醇酶、蘋果酸脫氫酶、泛素連接酶表達(dá)升高,己糖激酶表達(dá)下降。結(jié)論干酪乳酸桿菌代謝物對(duì)體外白假絲酵母菌生物被膜具有抑制作用。
[Abstract]:Objective to study the bacteriostatic effect of Lactobacillus casei metabolites on Candida albicans biofilm in vitro and its mechanism. Methods the metabolites of Lactobacillus casei were concentrated by extraction and distillation. The minimal inhibitory concentration (MIC80) of Lactobacillus casei metabolites on YEM30 free and biofilm forms of Candida albicans was determined by (XTT) reduction method. The effects of metabolites of Lactobacillus casei on the metabolism of Candida cerevisiae in vitro were quantitatively detected. The morphological changes of Candida albicans were observed and the differences of protein expression of Candida albicans before and after treatment with Lactobacillus casei metabolites were compared by 2-DE. Results the minimum inhibitory dilution times of the metabolites of Lactobacillus casei against Candida albicans and biofilm were 1: 256 and 1: 128 respectively. The metabolites of Lactobacillus casei with dilution multiple of 1: 256 could obviously interfere with the transformation from Candida albicans to hyphae. Six prominent protein spots were obtained from two-dimensional electrophoresis, and 4 of them were identified. The results showed that the expression of enolase, malate dehydrogenase and ubiquitin ligase increased under the stimulation of the metabolites of Lactobacillus casei. The expression of hexokinase decreased. Conclusion the metabolites of Lactobacillus casei can inhibit the biofilm of Candida albicans in vitro.
