【摘要】：目的比較目前常用的基因自發(fā)突變2型糖尿病小鼠C57BL/6J-db/db和STZ誘導(dǎo)的C57BL/6J小鼠的潰瘍創(chuàng)面愈合特征,為應(yīng)用穩(wěn)定的糖尿病小鼠潰瘍模型進(jìn)行相關(guān)動(dòng)物實(shí)驗(yàn)研究提供依據(jù)。方法建立糖尿病小鼠潰瘍模型,統(tǒng)計(jì)0、3、5、7、10、14 d小鼠創(chuàng)面動(dòng)態(tài)愈合率及愈合時(shí)間;7、14 d取組織,HE染色和Masson染色,免疫組化(CD31和PCNA)觀察創(chuàng)面病理組織變化;熒光定量測定愈合相關(guān)因子collagen-IIIα、α-SMA的基因表達(dá)。結(jié)果基因突變db/db小鼠較STZ誘導(dǎo)小鼠的創(chuàng)面愈合時(shí)間顯著延遲,由(16.6±0.8)d延長至(20.2±1.3)d(P0.001);db/db組較STZ組肉芽組織生長緩慢,新生上皮長度不足、膠原沉積紊亂,創(chuàng)面愈合較差;7 d,db/db組的CD31和PCNA顯著性低表達(dá)(P0.01);7、14 d,db/db組基因上調(diào)量的倍數(shù)明顯低于STZ組。結(jié)論兩種糖尿病小鼠創(chuàng)面均出現(xiàn)愈合延遲,但基因突變糖尿病db/db小鼠相比于STZ誘導(dǎo)組小鼠在創(chuàng)面愈合延遲的程度上和愈合難易程度上,更適合進(jìn)行糖尿病潰瘍創(chuàng)面的研究。
[Abstract]:Objective to compare the wound healing characteristics of C57BL/6J mice induced by C57BL/6J-db/db and STZ in type 2 diabetic mice with gene spontaneous mutation. Methods the ulcer model of diabetic mice was established. The dynamic wound healing rate and healing time were calculated on day 1014. The tissue was stained with HE and Masson. The pathological changes of wound were observed by immunohistochemistry (CD31 and PCNA). The gene expression of collagen-III 偽, 偽 -SMA was measured quantitatively. Results the wound healing time of db/db mice induced by STZ was significantly delayed than that of STZ mice. The wound healing time was prolonged from (16.6 鹵0.8) days to (20.2 鹵1.3) d (P0.001) d in db/db mice, and the granulation tissue grew slower, the length of neonate epithelium was insufficient and collagen deposition was disorder in STZ group than in STZ group. The expression of CD31 and PCNA was significantly lower (P0.01) in the group of poor wound healing at 7 days than in the group of STZ on the 7th day after treatment (P0.01), and the multiple of up-regulation of the gene was significantly lower in the group of STZ than that in the group of STZ. Conclusion both diabetic mice have delayed wound healing, but db/db mice with gene mutation are more suitable for the study of diabetic ulcer wounds than those in STZ induced mice.
