【摘要】：目的:考察補(bǔ)腎活血化痰方對(duì)多囊卵巢綜合征(PCOS)模型大鼠的治療作用機(jī)制。方法:將50只大鼠隨機(jī)分為正常對(duì)照組、模型對(duì)照組、陽性對(duì)照組[羅格列酮,3.0 mg/(kg·d)]和補(bǔ)腎活血化痰方高、低劑量組[7.5、5.0 g/(kg·d)],每組10只。除正常對(duì)照組外,其余各組大鼠均采用Poresky法復(fù)制PCOS模型。造模后,正常對(duì)照組和模型對(duì)照組大鼠ig生理鹽水,各給藥組大鼠ig相應(yīng)藥液,每天1次,連續(xù)22 d。給藥結(jié)束后,測(cè)定大鼠血清中睪酮(T)、抗苗勒氏管激素(AMH)、胰島素樣生長因子Ⅰ(IGF-Ⅰ)、腫瘤壞死因子α(TNF-α)水平和卵巢組織中轉(zhuǎn)化生長因子β1(TGF-β_1)陽性表達(dá)情況。結(jié)果:與正常對(duì)照組比較,模型對(duì)照組大鼠血清中T、AMH、IGF-Ⅰ、TNF-α水平及卵巢組織中TGF-β_1陽性表達(dá)率均顯著升高(P0.05);與模型對(duì)照組比較,各給藥組大鼠血清中T、AMH、IGF-Ⅰ、TNF-α水平及卵巢組織中TGF-β_1陽性表達(dá)率均顯著降低(P0.05),且補(bǔ)腎活血化痰方各劑量組指標(biāo)優(yōu)于陽性對(duì)照組(P0.05)。結(jié)論:補(bǔ)腎活血化痰方能明顯降低PCOS模型大鼠血清中T、AMH、IGF-Ⅰ、TNF-α水平以及卵巢組織中TGF-β_1陽性表達(dá)率,這可能是其治療PCOS的作用機(jī)制之一。
