BMP-2誘導(dǎo)低氧環(huán)境下BMSCs向軟骨表型分化

【摘要】:目的:探討B(tài)MP-2聯(lián)合低氧環(huán)境誘導(dǎo)BMSCs向成軟骨生成分化的可行性,并進(jìn)一步研究其生物學(xué)機(jī)制。 方法:體外培養(yǎng)4周齡健康清潔級雌性SD大鼠BMSCs,取第2代細(xì)胞根據(jù)培養(yǎng)條件不同將實(shí)驗(yàn)分為4組:常氧對照組(A組)、常氧加BMP-2誘導(dǎo)液組(B組)、低氧對照組(C組)和低氧加BMP-2誘導(dǎo)液組(D組)。倒置顯微鏡下觀察細(xì)胞形態(tài)變化,阿利新藍(lán)染色檢測各組軟骨基質(zhì)糖胺聚糖(glycosaminoglycans,GAG)分泌水平,Western blot檢測細(xì)胞內(nèi)Ⅱ型膠原和低氧誘導(dǎo)因子1α(hypoxia-inducible factor-1α,HIF-1α)蛋白表達(dá)水平,RT-PCR檢測成軟骨、成骨以及低氧相關(guān)基因表達(dá)水平。 結(jié)果:D組誘導(dǎo)21天,細(xì)胞變?yōu)轭悎A形,細(xì)胞密度明顯減少,細(xì)胞周邊呈陷窩樣,基質(zhì)包裹細(xì)胞,而A、B和C組均未見上述典型變化。D組阿利新藍(lán)染色較其他組明顯深染,并隨誘導(dǎo)時(shí)間延長,藍(lán)染加深,21天時(shí)出現(xiàn)成片深染藍(lán)色,其他實(shí)驗(yàn)組各時(shí)間點(diǎn)僅見散在少量的淡染藍(lán)色。Western Blot檢測細(xì)胞內(nèi)Ⅱ型膠原蛋白表達(dá)水平D組較其他實(shí)驗(yàn)組顯著增高(P0.05),HIF-1α蛋白表達(dá)水平C、D組較A、B組顯著增高,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。RT-PCR檢測D組成軟骨分化相關(guān)指標(biāo)COL2α1、Agg表達(dá)最高,而B組成骨分化相關(guān)指標(biāo)COL1α1、ALP、RUNX2表達(dá)水平最高,C和D組低氧相關(guān)的指標(biāo)HIF-1α較A和B組顯著增強(qiáng)(P0.05)。 結(jié)論:BMP-2聯(lián)合低氧環(huán)境可以很好地誘導(dǎo)BMSCs向軟骨表型分化,并抑制其成骨表型分化,,HIF-1α是參與其促軟骨生成過程可能機(jī)制的一個(gè)重要信號分子。

Methods: the second passage of BMSCs, cells of healthy clean grade female SD rats cultured in vitro were divided into four groups: normoxic control group (group A), normoxic plus BMP-2 induction solution group (group B) and hypoxic control group (group C).
