【摘要】：Pyroptosis是最近發(fā)現(xiàn)的一種依賴于半胱氨酸蛋白-1(Caspase-1)的程序性細胞死亡方式。這種死亡方式和細胞凋亡及其它程序性死亡在細胞學(xué)形態(tài)、分子機制上有著本質(zhì)的區(qū)別。其主要特征為依賴于Caspase-1剪切激活和促炎癥性細胞因子白介素(IL)-1β和IL-18的釋放。而Caspase-1的活化受到胞內(nèi)蛋白復(fù)合物炎癥體的調(diào)控。該炎癥體由NLRs蛋白(如NLRP3或NLRC4)、接頭蛋白ASC以及Caspase-1構(gòu)成。Pyroptosis最初是在傷寒沙門氏菌感染小鼠巨噬細胞的模型中被發(fā)現(xiàn),隨后研究顯示Pyroptosis在細菌和病毒感染、自身免疫性疾病以及腫瘤發(fā)生中都起到關(guān)鍵性作用。對這種程序性死亡的深入研究將有助于對細菌感染防治和某些疾病的致病機制研究提供幫助,并最終為臨床應(yīng)用提供新思路。
[Abstract]:Pyroptosis is a recently discovered programmed cell death pattern dependent on cysteine protein 1 (Caspase-1). This mode of death is essentially different from apoptosis and other programmed cell death in cellular morphology and molecular mechanism. Its main characteristics are dependent on the activation of Caspase-1 shear and the release of interleukin (IL)-1 尾 and IL-18. The activation of Caspase-1 is regulated by the inflammatory body of intracellular protein complex. The inflammatory body consisted of NLRs proteins (such as NLRP3 or NLRC4), junction protein ASC, and Caspase-1. Pyroptosis was first found in a murine macrophage model infected with Salmonella typhimurium. Later studies showed that Pyroptosis was infected by bacteria and viruses. Autoimmune diseases and tumorigenesis play a key role. The further study of this kind of programmed death will be helpful to the prevention and treatment of bacterial infection and the study of pathogenetic mechanism of some diseases, and will finally provide a new idea for clinical application.
