酸適應(yīng)海德爾堡沙門氏菌對環(huán)境脅迫耐受性分析

【摘要】:目的:研究海德爾堡沙門氏菌(Salmonella heidelberg)低pH值酸適應(yīng)性及酸適應(yīng)對細(xì)胞其他脅迫環(huán)境耐受性的影響。方法:海德爾堡沙門氏菌在pH 5.5酸性培養(yǎng)基中生長2h誘導(dǎo)酸適應(yīng)性,以pH 3.0酸脅迫培養(yǎng)基及不同溫度、NaCl、膽鹽、消毒劑環(huán)境的存活率評(píng)價(jià)其耐受性。結(jié)果:pH 5.5酸適應(yīng)2 h后,海德爾堡沙門氏菌在pH 3.0酸脅迫環(huán)境的存活率大幅提高。低pH值酸適應(yīng)也使海德爾堡沙門氏菌在45、50℃高溫條件下的存活率顯著提高,對4℃冷藏環(huán)境的耐受性顯著低于非酸處理菌株,但對-20℃冷凍環(huán)境的生存無影響。酸適應(yīng)海德爾堡沙門氏菌對高滲、膽鹽脅迫也表現(xiàn)良好的耐受性,5~10 g/100mL NaCl溶液處理1 h,酸適應(yīng)海德爾堡沙門氏菌的存活率大于60%,在1~10 g/100mL膽鹽溶液的存活率比對照組高3.8倍。海德爾堡沙門氏菌酸適應(yīng)性也使其對消毒劑的敏感性發(fā)生變化,酸適應(yīng)性海德爾堡沙門氏菌在10~25 mmol/LH_2O_2溶液的存活率顯著高于對照菌株,最高達(dá)3.1倍;但對NaClO、乙醇消毒劑的耐受性顯著降低,高質(zhì)量濃度下二者無顯著差異。結(jié)論:海德爾堡沙門氏菌在低pH值酸性條件生長一段時(shí)間會(huì)誘導(dǎo)產(chǎn)生酸適應(yīng)性,還會(huì)協(xié)同提高其對高溫、高滲、膽鹽、H_2O_2脅迫環(huán)境的交叉抗性;但對4、-20℃低溫、NaClO、乙醇等殺菌、保藏措施敏感性提高。
