脂肪干細胞治療促進組織血管化的旁分泌機制中細胞因子來源的探討

【摘要】:目的:探索脂肪干細胞(adipose-derived cells,ADSCs)治療促進組織血管化的治療機理,為細胞治療的臨床轉化研究提供理論基礎。 方法:1、ADSCs的分離培養(yǎng)與鑒定 在無菌條件下,取出wistar大鼠腹股溝脂肪墊,用氯霉素沖洗,于膠原酶中振蕩消化,低速離心,細胞篩網過濾后接種于培養(yǎng)皿,獲得原代ADSCs。 取第三代ADSCs用成軟骨、成脂誘導培養(yǎng)液誘導培養(yǎng),,分別行Ⅱ型膠原染色及油紅O染色鑒定。 2、建立裸鼠隨意皮瓣模型 在裸鼠背部掀起1cm×2.5cm大小隨意皮瓣,于皮瓣肉膜面分別注射約1×107個已標記CM-Dil的ADSCs0.2ml混懸液(實驗組)及等體積的PBS(對照組)。 3、血管化細胞因子水平分析 3、1將植入的ADSCs與裸鼠自身的細胞分選出 上述動物模型一部分分別在術后第1、3、7天取材,將皮瓣組織于無菌條件下取出并剪碎,用0.2%的膠原酶于37℃振蕩消化1h后,用等體積含10%胎牛血清DMEM液中止消化,離心后用2%FBS重懸,用流式細胞儀將DiL標記的陽性細胞與裸鼠自身的陰性細胞分選出。 3、2分選后的細胞提RNA行VEGF、bFGF及HIF-1α的定量PCR分析。 結果:1、大鼠ADSCs貼壁后呈成纖維細胞樣形態(tài)。第三代ADSCs經成脂、成軟骨誘導分化后,染色結果均為陽性。 2、ADSCs治療組皮瓣成活率顯著高于對照組,p0.001,有統(tǒng)計學差異。 3、流式細胞分選結果提示,在細胞治療后第1、3、7天,植入的細胞數(shù)量(帶熒光標記的陽性細胞)呈明顯下降趨勢。 4、定量PCR結果提示:經細胞治療干預的皮瓣組織,其HIF-1α及其下游分子的表達水平較對照組高,且隨時間推移有增加趨勢。 結論:促進損傷組織血管化的細胞因子來源于裸鼠自身,而并非植入的ADSCs所分泌。證實了細胞的植入其實是通過上調組織自身的血管化因子水平,來間接促進組織血管新生的。
