血清放置時(shí)間對(duì)大鼠血生化檢測(cè)的影響

【摘要】:目的探討大鼠血清放置時(shí)間對(duì)14項(xiàng)血生化檢測(cè)結(jié)果的影響。方法隨機(jī)選取40只成年SD大鼠,空腹采集靜脈血液標(biāo)本,靜置1 h,分離血清并密封分裝,存放于4℃和-20℃冰箱內(nèi),應(yīng)用全自動(dòng)生化分析儀于分離血清后0 h、4 h、24 h、96 h、7 d檢測(cè)谷丙轉(zhuǎn)氨酶(ALT)、谷草轉(zhuǎn)氨酶(AST)、堿性磷酸酶(ALP)、總蛋白(TP)、白蛋白(ALB)、肌酐(CREA-J)、尿酸(UA)、尿素氮(UREA)、血糖(GLU)、總膽固醇(TC)、甘油三酯(TG)、低密度脂蛋白膽固醇(LDL-C)、肌酸激酶(CK)和乳酸脫氫酶(LDH)等14項(xiàng)血生化指標(biāo),并進(jìn)行比較。結(jié)果雌雄大鼠血生化數(shù)據(jù)變化趨勢(shì)一致。4℃保存時(shí),與0 h相比,ALP在4 h、24 h、96 h、7d均顯著降低(P0.05),ALB在96 h和7 d均極顯著升高(P0.01),CREA-J在96 h和7 d均顯著升高(P0.05),UA在24 h、96 h、7 d均極顯著升高(P0.01),其他指標(biāo)差異無(wú)顯著性(P0.05)。-20℃保存時(shí),與0 h相比,ALT在7 d顯著升高(P0.01),AST在96 h和7 d顯著升高(P0.05),TP在4 h和24 h顯著降低(P0.05),ALB在4 h、24 h、96 h、7 d均顯著升高(P0.01),CREA-J在24 h、96 h、7 d均顯著升高(P0.01),UA在4 h、24 h、96 h、7 d均顯著升高(P0.01),TC在4 h、24 h、96 h、7 d均顯著升高(P0.05),TG在96 h和7 d顯著升高(P0.05),CK在96 h和7 d顯著升高(P0.05),LDH在96 h和7 d顯著升高(P0.05),其他指標(biāo)差異無(wú)顯著性(P0.05)。結(jié)論大鼠血生化測(cè)定盡可能在采集血清后即刻完成,尤其是ALP的測(cè)定,4℃冷藏時(shí),測(cè)定UA不宜超過(guò)4 h,測(cè)定ALB和CREA-J不宜超過(guò)24 h,測(cè)定ALT、AST、TP、UREA、GLU、TC、TG、LDL-C、CK及LDH等指標(biāo)不宜超過(guò)7 d。
