忍冬藤顆粒對急性痛風性關(guān)節(jié)炎模型大鼠形態(tài)學及關(guān)節(jié)軟骨IL-1β的影響

【摘要】:目的:研究忍冬藤顆粒治療急性痛風性關(guān)節(jié)炎模型大鼠抗炎消腫止痛的效果并對其機理進行探討。方法:給予大鼠乙胺丁醇(189mg/kg)和腺嘌呤(1.26g/kg)灌胃,每天1次連續(xù)3周誘導高尿酸血癥模型,然后在此基礎(chǔ)上以大鼠的右踝關(guān)節(jié)外側(cè)后方為穿刺點,針尖斜向前上方與脛骨成45°角刺入關(guān)節(jié)腔,每只注射0.2ml尿酸鈉溶液,連續(xù)3天。同時各干預組大鼠分別灌胃忍冬藤顆粒23.8g/kg、11.9g/kg、5.95g/kg,別嘌呤醇0.455g/kg(第3周開始,連續(xù)1周),秋水仙堿18.9mg/kg(第4周開始.連續(xù)1周)進行干預。實驗結(jié)束后,觀察忍冬藤顆粒對模型大鼠關(guān)節(jié)腫脹、步態(tài)、炎癥的影響,檢測大鼠關(guān)節(jié)軟骨中的IL-1β的表達水平。結(jié)果:與空白組比較,模型組大鼠關(guān)節(jié)腫脹增加(6.20±0.19),關(guān)節(jié)炎癥(5.25±1.04)、跛行步態(tài)評分(5.25±1.04)增高,關(guān)節(jié)軟骨IL-1β表達水平上升(12.33±1.36);與模型組比較,藥物干預24h、48h、72h后,忍冬藤顆粒各組大鼠關(guān)節(jié)腫脹度周徑明顯減小,其中忍冬藤顆粒11.9g/kg劑量組對關(guān)節(jié)腫脹度的療效更好(4.51±0.25);與模型組比較,給藥后72h后,秋水仙堿(18.9mg/kg)+別嘌呤醇(0.455g/kg)組可以明顯降低關(guān)節(jié)炎癥程度,給藥48、72h后,忍冬藤各劑量組可以明顯降低關(guān)節(jié)炎癥評分,其中忍冬藤顆粒11.9g/kg劑量組降低炎癥評分效果更優(yōu)(1.00±1.07);與模型組比較,給藥后24h、48h、72h后,忍冬藤顆粒各劑量組可以明顯降低步態(tài)評分(1.00±1.07);與模型組比較,秋水仙堿(18.9mg/kg)+別嘌呤醇(0.455g/kg)、忍冬藤顆粒各劑量組大鼠血清IL-1β表達明顯降低,其中忍冬藤顆粒11.9g/kg(6.16±0.70)、23.8g/kg(6.96±0.79)劑量組IL-1β表達降低效果最優(yōu)。結(jié)論:忍冬藤顆粒可以起到抗炎消腫止痛的功效,對急性痛風性關(guān)節(jié)炎模型大鼠有著明顯的治療作用,其作用機制可能是抑制炎癥關(guān)節(jié)IL-1β致炎因子的生成而實現(xiàn)的。
[Abstract]:Objective: to study the effect and mechanism of Lonicera japonicus granule in treating acute gouty arthritis model rats. Methods: rats were given ethambutanol (189mg/kg) and adenine (1.26g/kg) intragastrically to induce hyperuricemia model once a day for 3 weeks. A 45 擄angle between the tip oblique and the tibia was inserted into the articular cavity. Each mouse was injected with sodium 0.2ml uric acid for 3 days. At the same time, the rats in each intervention group were treated by intragastric administration of Lonicera japonicus granules 23.8g / kg 11.9g / kg, allopurinol 0.455g/kg 5.95g / kg, and colchicine 18.9mg/kg at the beginning of week 3, respectively, and the rats were given colchicine 18.9mg/kg (from week 4). Intervention was carried out for one week in a row. At the end of the experiment, the effects of Lonicera japonicus granule on joint swelling, gait and inflammation were observed, and the expression level of IL-1 尾 in articular cartilage of rats was detected. Results: compared with the blank group, the joint swelling increased (6.20 鹵0.19), arthritis (5.25 鹵1.04), limping gait score (5.25 鹵1.04) and articular cartilage IL-1 尾 expression increased (12.33 鹵1.36) in the model group. The radius of swelling degree of joint decreased obviously in each group, and the effect of 11.9g/kg group was better (4.51 鹵0.25), compared with the model group, 72 hours after administration, the effect was better than that in the model group, and compared with the model group, after 72 hours of administration, the effect of 11.9g/kg on the degree of joint swelling was better than that in the model group. Colchicine (18.9mg/kg) allopurinol (0.455g/kg) group could significantly reduce the degree of arthritis. Compared with the model group, each dose group of Lonicera Lonicera granule at 48 hours and 72 hours after administration significantly decreased the gait score (1.00 鹵1.07), compared with the model group, and compared with the model group, each dose group of Lonicera Lonicera granule decreased the score of gait obviously (1.00 鹵1.07), compared with the model group. The expression of serum IL-1 尾 in the colchicine (18.9mg/kg) allopurinol (0.455g/kg) and Lonicera japonica granule groups was significantly decreased, and the decrease of IL-1 尾 expression was the best in the group of 11.9g/kg (6.16 鹵0.70) and 23.8g / kg (6.96 鹵0.79). Conclusion: Lonicera japonicus granule can inhibit inflammation, detumescence and pain, and has obvious therapeutic effect on acute gouty arthritis rats, and its mechanism may be to inhibit the production of IL-1 尾 inflammatory factor in inflammatory joints.
