【摘要】：目的探討纖維蛋白原樣2凝血酶原酶(FGL2)在急性病毒感染過程中對(duì)T細(xì)胞的增殖、分化和功能的影響。方法模型一:淋巴細(xì)胞脈絡(luò)叢腦膜炎病毒(LCMV)感染Fgl2基因敲除小鼠(實(shí)驗(yàn)組)與C57BL/6J對(duì)照小鼠,感染后第8天,檢測(cè)T細(xì)胞的增殖、分化和功能。模型二:過繼轉(zhuǎn)移能夠特異性識(shí)別LCMV的TCR轉(zhuǎn)基因小鼠SMARTA或P14的T細(xì)胞到Fgl2基因敲除小鼠(實(shí)驗(yàn)組)和C57BL/6J對(duì)照小鼠內(nèi),LCMV感染后第8天,檢測(cè)小鼠體內(nèi)供體SMARTA或P14細(xì)胞的增殖和分化。結(jié)果FGL2在LCMV急性感染過程中表達(dá)上調(diào)(P0.05);實(shí)驗(yàn)組和對(duì)照組的濾泡輔助T細(xì)胞(T_(FH))、Th1細(xì)胞和效應(yīng)CD8~+T細(xì)胞的頻率和數(shù)量均無(wú)明顯差異(P0.05);GC B細(xì)胞和漿細(xì)胞的數(shù)量和頻率、LCMV特異性抗體的滴度以及效應(yīng)CD8~+T細(xì)胞中表達(dá)TNF-α和CD107a/b的頻率均無(wú)明顯差異(P0.05)。結(jié)論 FGL2在急性病毒感染過程中不影響T細(xì)胞的免疫應(yīng)答。
[Abstract]:Objective to investigate the effects of fibrinogen 2 prothrombin (FGL2) on the proliferation, differentiation and function of T cells during acute viral infection. Methods: model 1: Fgl2 gene knockout mice (experimental group) and C57BL/6J control mice infected with lymphocytic choroid plexitis virus (LCMV) were tested for T cell proliferation, differentiation and function on the 8th day after infection. Model 2: adoptive transfer could specifically recognize the SMARTA or P14 T cells of TCR transgenic mice with LCMV to Fgl2 knockout mice (experimental group) and C57BL/6J control mice on the 8th day after LCMV infection. The proliferation and differentiation of donor SMARTA or P14 cells in mice were detected. Results the expression of FGL2 was up-regulated during acute LCMV infection (P0.05), and there was no significant difference in the frequency and number of T _ (FH) T cells (T _ (FH) Th1) and effector CD8~ T cells between the experimental group and the control group (P0.05). There was no significant difference in the titer of heterosexual antibodies and the frequency of expression of TNF- 偽 and CD107a/b in CD8~ T cells (P0.05). Conclusion FGL2 does not affect the immune response of T cells during acute viral infection.
