抗B7-H3單克隆抗體抑制雨蛙肽誘導(dǎo)的小鼠急性胰腺炎炎癥反應(yīng)和組織損傷

【摘要】:目的探究抗B7-H3單克隆抗體(m Ab)在雨蛙肽誘導(dǎo)的急性胰腺炎(AP)中的作用。方法將小鼠隨機(jī)分為對照組、AP組、抗B7-H3 m Ab治療組。采用腹腔注射雨蛙肽的方法制備小鼠AP模型,治療組于造模前1 h皮下注射抗B7-H3 m Ab,于造模后6、12、24 h,收集各組小鼠血液、胰腺及肺組織。采用Western blot法和免疫組織化學(xué)染色檢測對照組及AP組小鼠胰腺組織B7-H3蛋白表達(dá)。應(yīng)用全自動生化免疫分析儀檢測各組小鼠血清淀粉酶及脂肪酶活性;胰腺組織濕干比評估各組小鼠胰腺組織水腫變化;HE染色評估各組小鼠胰腺及肺組織病理變化,利用ELISA檢測各組小鼠血清中腫瘤壞死因子α(TNF-α)、白細(xì)胞介素6(IL-6)、IL-1β水平。結(jié)果 B7-H3蛋白在AP模型小鼠胰腺組織高表達(dá),且在12 h左右達(dá)到高峰;AP組血清淀粉酶及脂肪酶明顯高于對照組,抗B7-H3 m Ab處理后明顯降低;AP組胰腺及肺組織出現(xiàn)明顯炎癥反應(yīng),治療組炎癥反應(yīng)明顯減弱,胰腺組織濕干比大幅降低;AP組TNF-α、IL-6、IL-1β水平隨時間延長而升高,12 h達(dá)到高峰后開始下降,而治療組促炎因子水平相對降低。結(jié)論 B7-H3在雨蛙肽誘導(dǎo)的AP模型中存在高表達(dá),抗B7-H3 m Ab可以減弱炎癥反應(yīng),緩解胰腺及肺組織的損傷。
[Abstract]:Objective to investigate the role of anti-B7-H3 monoclonal antibody (m Ab) in acute pancreatitis (AP) induced by rainfrog peptide. Methods mice were randomly divided into control group (AP group) and anti B7-H3 m Ab group. The AP model of mice was established by intraperitoneal injection of rain frog peptide. In the treatment group, anti B7-H3 m Ab, was injected subcutaneously 1 hour before the model was made. The blood, pancreas and lung tissues of each group were collected. The expression of B7-H3 protein in pancreas of control group and AP group was detected by Western blot and immunohistochemical staining. The activities of serum amylase and lipase in each group were detected by automatic biochemical immunoassay, the changes of pancreatic edema in each group were evaluated by wet / dry ratio of pancreatic tissue and the pathological changes of pancreas and lung in each group were evaluated by HE staining. Serum levels of tumor necrosis factor 偽 (TNF- 偽) and interleukin-6 (IL-6) IL-1 尾 were measured by ELISA. Results the expression of B7-H3 protein in pancreatic tissue of AP model mice was high, and the levels of serum amylase and lipase in AP group were significantly higher than those in control group at about 12 h, and the inflammatory reaction in pancreas and lung tissue of AP group was obviously decreased after treatment with anti-B7-H3 m Ab. In the treatment group, the inflammatory response was obviously weakened, the wet / dry ratio of pancreatic tissue was significantly decreased, and the level of TNF- 偽, IL-6 and IL-1 尾 in the AP group increased with the prolongation of time and reached its peak at 12 h, but the level of proinflammatory factor decreased relatively in the treatment group. Conclusion there is a high expression of B7-H3 in the AP model induced by rain frog peptide. Anti B7-H3 m Ab can attenuate the inflammatory reaction and alleviate the injury of pancreas and lung tissue.
