【摘要】：目的:通過對比不同方法處理COPD模型大鼠肺組織,尋找適當(dāng)方法減少在COPD模型大鼠細(xì)胞培養(yǎng)中的污染機(jī)會,提高細(xì)胞培養(yǎng)成功率。方法:將同一COPD模型大鼠肺組織分成兩部分,分別用不同方法對其預(yù)處理,用組織塊培養(yǎng)法進(jìn)行原代細(xì)胞培養(yǎng),觀察第24h、72h及后續(xù)培養(yǎng)情況。結(jié)果:經(jīng)過刮除肺葉及氣管內(nèi)膜的組織原代細(xì)胞培養(yǎng)組污染率較直接組織培養(yǎng)的要低,肺葉培養(yǎng)污染率為100%。結(jié)論:(1)刮除肺葉及氣管內(nèi)膜的方法有助于減少COPD模型大鼠原代細(xì)胞培養(yǎng)組織法的污染;(2)COPD模型大鼠肺成纖維細(xì)胞原代培養(yǎng)的污染來源主要為肺部感染灶。
[Abstract]:Aim: to study the effects of different methods on the treatment of lung tissue in COPD model rats, and to find appropriate ways to reduce the chance of contamination in COPD model rats' cell culture and to improve the success rate of cell culture. Methods: the lung tissues of the same COPD model rats were divided into two parts. The primary cells were cultured by tissue mass culture and the 24 h / 72 h and subsequent culture were observed. Results: the contamination rate of primary cell culture group was lower than that of direct tissue culture group, and the contamination rate of lung lobe culture was 100%. Conclusion: (1) the method of scraping the lung lobe and endotracheal intima is helpful to reduce the pollution of primary cell culture organic law in COPD model rats, and (2) the primary source of contamination of lung fibroblasts in COPD model rats is mainly pulmonary infection.
