縫隙連接蛋白43和黃體生成素受體在小鼠卵巢中的表達(dá)

【摘要】:目的觀察小鼠卵泡發(fā)育過程中縫隙連接蛋白43(Cx43)和黃體生成素受體(LHR)在小鼠卵巢中的表達(dá)以及黃體生成素(LH)對(duì)卵巢Cx43表達(dá)的影響,為這兩種分子與卵泡發(fā)育的關(guān)系提供實(shí)驗(yàn)依據(jù)。方法選擇出生后1d、4d、7d、2周、3周、4周、6周、8周齡雌性C57小鼠共8組,每組10只,取卵巢。HE染色觀察卵巢形態(tài)、卵泡發(fā)育并測(cè)量顆粒細(xì)胞體積分?jǐn)?shù)。免疫組織化學(xué)和Western blotting觀察卵巢組織中Cx43及LHR的表達(dá),Western blotting檢測(cè)不同濃度黃體生成素(LH)孵育后對(duì)卵巢Cx43表達(dá)的影響。結(jié)果 HE染色顯示,出生1d小鼠卵巢中見原始卵泡,出生7d卵巢內(nèi)出現(xiàn)初級(jí)卵泡,2周時(shí)出現(xiàn)竇前卵泡和少量竇卵泡,3周、4周時(shí),出現(xiàn)較多大的竇卵泡,6~8周時(shí)出現(xiàn)黃體。卵泡顆粒細(xì)胞體積分?jǐn)?shù)在2周后的卵巢中明顯增加。Cx43表達(dá)在各階段卵泡的顆粒細(xì)胞胞膜上,LHR表達(dá)在卵泡膜細(xì)胞、顆粒細(xì)胞及卵母細(xì)胞胞質(zhì)中。Cx43和LHR出生后2周卵巢中的表達(dá)開始明顯增加,并隨卵泡發(fā)育表達(dá)逐漸增強(qiáng),在3周、4周、6周和8周維持相對(duì)較高水平。LH體外干預(yù)可增加卵巢組織Cx43的表達(dá)。結(jié)論 Cx43和LHR在卵泡發(fā)育中發(fā)揮重要作用,LH可能通過LHR促進(jìn)卵巢卵泡顆粒細(xì)胞Cx43的表達(dá)。
[Abstract]:Objective to observe the expression of gap junction protein 43 (Cx43) and luteinizing hormone receptor (LHR) in mouse ovary during follicular development and the effect of luteinizing hormone (LH) on the expression of Cx43 in ovary, and to provide experimental evidence for the relationship between these two molecules and follicular development. Methods A total of 10 female C57 mice (10 in each group) were divided into 8 groups, including 10 female C57 mice at the age of 8 weeks, 1 day and 4 days and 7 days after birth. Ovarian morphology, follicle development and granulosa cell volume fraction were observed with HE staining. Immunohistochemistry and Western blotting were used to detect the expression of Cx43 and LHR in ovarian tissue. Western blotting was used to detect the effect of different concentrations of luteinizing hormone (LH) on the expression of Cx43 in ovary. Results HE staining showed that primordial follicles were found in the ovaries of 1 day old mice, preantral follicles were found in the ovary of 7 days after birth, preantral follicles appeared in 2 weeks and a few antral follicles appeared in 4 weeks, and more large antral follicles appeared in corpus luteum at 68 weeks after birth. The volume fraction of follicular granulosa cells in the ovary increased significantly after 2 weeks. Cx43 was expressed on the membrane of granulosa cells in all stages of follicles, and LHR was expressed in follicular membrane cells. The expression of. Cx43 and LHR in the cytoplasm of granulosa cells and oocytes began to increase 2 weeks after birth, and increased gradually with follicular development. At 3 weeks, 4 weeks, 6 weeks and 8 weeks, the level of LH was relatively high. LH increased the expression of Cx43 in ovarian tissues. Conclusion Cx43 and LHR play an important role in follicular development. LH may promote the expression of Cx43 in follicular granulosa cells through LHR.
