芎歸六君子湯對慢性阻塞性肺疾病氣虛血瘀痰阻證模型大鼠氣道Muc5ac水平的影響

【摘要】:目的觀察芎歸六君子湯對慢性阻塞性肺疾病(COPD)氣虛血瘀痰阻證模型大鼠氣道Muc5ac水平的影響。方法60只大鼠按隨機數(shù)字表隨機分為空白組、芎歸六君子湯自身對照組、模型組、芎歸六君子湯干預(yù)組(低、中、高劑量),每組10只。采用改良的香煙煙熏和脂多糖氣管注入法聯(lián)合制備模型。采用芎歸六君子湯干預(yù)15天后取材,觀察肺泡灌洗液(BALF)中Muc5ac水平、氣道上皮Muc5ac蛋白表達及肺組織Muc5ac mRNA表達。結(jié)果與空白組比較,模型組大鼠BALF中的Muc5ac水平及氣道上皮Muc5ac蛋白的表達增加,肺組織Muc5ac mRNA表達上調(diào)(P0.05)。芎歸六君子湯能降低COPD氣虛血瘀痰阻證模型大鼠BALF中的Muc5ac水平、氣道上皮Muc5ac蛋白表達,下調(diào)肺組織Muc5ac mRNA表達,且以高劑量組較為明顯(P0.05)。結(jié)論芎歸六君子湯在一定程度上降低COPD氣虛血瘀痰阻證模型大鼠氣道Muc5ac水平,減少氣道黏液高分泌。
[Abstract]:Objective to observe the effect of Xionggui Liujunzi decoction on the level of airway Muc5ac in (COPD) rats with Qi deficiency and blood stasis and phlegm blocking syndrome. Methods Sixty rats were randomly divided into blank group, control group, model group and intervention group (low, medium and high dosage) with 10 rats in each group. The model was prepared by modified cigarette fumigation and endotracheal injection of lipopolysaccharide (LPS). After treated with Xionggui Liujunzi decoction for 15 days, the levels of Muc5ac in alveolar lavage fluid (BALF), the expression of Muc5ac protein in airway epithelium and the expression of Muc5ac mRNA in lung tissue were observed. Results compared with the blank group, the level of Muc5ac in BALF and the expression of Muc5ac protein in airway epithelium were increased in the model group, and the expression of Muc5ac mRNA was up-regulated in the lung tissue (P0.05). Xionggui Liujunzi decoction could reduce the level of Muc5ac in BALF, Muc5ac protein expression in airway epithelium and Muc5ac mRNA expression in lung tissue of COPD rats with Qi deficiency and blood stasis and phlegm obstruction, especially in high dose group (P0.05). Conclusion Xionggui Liujunzi decoction can reduce the level of airway Muc5ac and reduce airway mucus hypersecretion in COPD rats with Qi deficiency and blood stasis and phlegm obstruction to some extent.
