丹參酮對(duì)人工關(guān)節(jié)無菌性松動(dòng)小鼠氣囊模型中炎癥反應(yīng)的影響

【摘要】:目的:研究丹參酮對(duì)人工關(guān)節(jié)無菌性松動(dòng)小鼠氣囊模型中炎癥反應(yīng)的影響。方法:將小鼠隨機(jī)分為空白對(duì)照組(生理鹽水)、鈦顆粒組(生理鹽水)和丹參酮低、中、高劑量組(50、100、200 mg/kg),每組10只。所有小鼠建立氣囊模型,除空白對(duì)照組小鼠氣囊內(nèi)注入0.5 mL生理鹽水外,其余各組小鼠氣囊內(nèi)均注入0.5 mL鈦顆粒懸液(10 mg/mL),24 h后按0.1 mL/10 g連續(xù)ig相應(yīng)藥物14 d。末次給藥24 h后收集氣囊,肉眼和蘇木精-伊紅染色顯微鏡觀察氣囊炎癥情況,計(jì)算炎癥細(xì)胞密度;實(shí)時(shí)定量聚合酶鏈?zhǔn)椒磻?yīng)法檢測(cè)氣囊組織中腫瘤壞死因子α(TNF-α)、白細(xì)胞介素1β(IL-1β)m RNA表達(dá)量;酶聯(lián)免疫吸附法檢測(cè)氣囊組織中TNF-α、IL-1β蛋白表達(dá)量。結(jié)果:與空白對(duì)照組比較,鈦顆粒組小鼠氣囊紅腫明顯,可見較多滲出及新生血管,炎癥反應(yīng)嚴(yán)重,炎癥細(xì)胞密度明顯增加(P0.05);TNF-α、IL-1βm RNA及蛋白表達(dá)明顯增強(qiáng)(P0.05)。與鈦顆粒組比較,丹參酮各劑量組小鼠氣囊紅腫減輕,滲出及新生血管減少,炎癥反應(yīng)減輕,炎癥細(xì)胞密度明顯減小(P0.05);TNF-α、IL-1βm RNA及蛋白表達(dá)明顯減弱(P0.05),且呈劑量依賴性。結(jié)論:丹參酮可有效抑制人工關(guān)節(jié)無菌性松動(dòng)小鼠氣囊模型中的無菌性炎癥反應(yīng)。
[Abstract]:Aim: to study the effect of tanshinone on inflammatory reaction in aseptic loosening mouse airbag model. Methods: the mice were randomly divided into control group (normal saline), titanium granule group (normal saline) and tanshinone low, medium and high dose group (50100200 mg/kg), each group had 10 rats. All the mice were injected with 0. 5 mL normal saline into the airbag of the control group, and 0. 5 mL titanium granule suspension (10 mg/mL) was injected into the airbag of all the mice for 24 h, and the corresponding drug was given orally for 14 days at the dose of 0. 1 mL/10 g. After the last administration for 24 hours, the air sacs were collected, and the inflammation was observed by naked eye and hematoxylin-eosin staining microscope, and the density of inflammatory cells was calculated. The expression of tumor necrosis factor- 偽 (TNF- 偽) and interleukin-1 尾 (IL-1 尾) m RNA) was detected by real-time quantitative polymerase chain reaction (RQPCR), and the expression of TNF- 偽 and IL-1 尾 was detected by enzyme-linked immunosorbent assay (Elisa). Results: compared with the blank control group, the mice in the titanium granule group showed obvious redness and swelling of the airbag, more exudation and neovascularization, severe inflammatory reaction, and obvious increase in the density of inflammatory cells (P0.05). The expression of IL-1 尾 m RNA and protein in the TNF- 偽 + IL-1 尾 m group was significantly increased (P0.05). Compared with the titanium granule group, the balloon redness, exudation and neovascularization, inflammatory reaction and the density of inflammatory cells decreased significantly in tanshinone group (P0.05), and the expression of IL-1 尾 m RNA and protein decreased significantly (P0.05) in a dose-dependent manner. Conclusion: tanshinone can effectively inhibit the aseptic inflammatory reaction in aseptic loosening mouse airbag model.
